Analysis of candidate genes underlying two epistatic quantitative trait loci on SSC12 affecting litter size in pig

P>The previous results from a genome scan for total number of piglets born and number of piglets born alive in a F-2 Iberian by Meishan intercross showed several single and epistatic QTL. One of the most interesting results was obtained for SSC12, where two QTL affecting both traits showed epistatic interaction. In this study, we proposed two genes (SLC9A3R1 and NOS2) as biological and potentially positional candidates underlying these QTL. Both cDNAs were characterized and 23 polymorphisms were detected. A chromosome scan was conducted with 12 markers, plus one SNP in SLC9A3R1 and one in NOS2, covering 110 cM of SSC12. The epistatic QTL (QTL1 at 15 cM and QTL2 at 97 cM) were confirmed, and SLC9A3R1 and NOS2 were mapped around the QTL1 and QTL2 regions respectively. Several SNPs in both genes were tested with standard animal model and marker assisted association tests. The most significant results were obtained with the NOS2 haplotype defined by one missense SNP c.2192C > T (Val to Ala) and a 15 bp duplication at the 3'UTR. This duplication seems to include AU-rich elements, and could be a target site for miRNA, therefore there are statistical and biological indications to consider this haplotype as the potential causal mutation underlying QTL2. SLC9A3R1 results were not conclusive. Although the interaction between the SNPs was not significant, we cannot reject the possibility of interaction of the NOS2 haplotype with other polymorphisms closely linked to the SL9A3R1 SNPs analysed.

Summary: The standard algorithm for diagnosing hepatitis C virus (HCV) infection involves an HCV antibody test for screening and a nucleic acid amplification test (NAAT) for confirmation. The accuracy of the Abbott ARCHITECT HCV Ag assay was evaluated in people living with HIV/AIDS (PLWHA), showing good performance in high-prevalence settings but poor in low-prevalence settings. It was excellent in excluding active HCV infection.

Relative telomere length impact on mortality of COVID-19: Sex differences

Summary: Increasing age is associated with severity and higher mortality of COVID-19. Telomere shortening is associated with higher risk of infections and may be used to identify those patients who are more likely to die. This study evaluated the association between relative telomere length (RTL) and COVID-19 mortality. Lower RTL was associated with a higher risk of death in women at 30 and 90 days.

Summary: This study assessed the effectiveness of a protein-based meningococcal vaccine in preventing invasive meningococcal disease in Spanish children. The results showed that complete vaccination with the vaccine was 76% effective against invasive meningococcal disease caused by any serogroup, and partial vaccination was 54% effective.

PBMCs gene expression signature of advanced cirrhosis with high risk for clinically significant portal hypertension in HIV/HCV coinfected patients: A cross-control study

Summary: We identified a gene expression signature associated with advanced cirrhosis with high risk for clinically significant portal hypertension (CSPH) in HIV/HCV coinfected patients through analyzing the gene expression profile of peripheral blood mononuclear cells (PBMCs). A cross-sectional study on 68 patients was conducted, and liver stiffness measurement (LSM) was used for stratification. We found 60 differentially expressed transcripts between patients with LSM < 12.5 kPa and >= 25 kPa, and two of them were validated in a different cohort.

Summary: It has been found that gene expression can be blocked by methylation during gametogenesis, resulting in unequal genetic contribution from parents to offspring. The study on a population of Purebred Spanish (PRE) horses revealed that both maternal and paternal gametic effects influence reproductive and morphological traits. The maternal gametic effect generally has a stronger influence on most traits.

Summary: This meta-analysis evaluated the diagnostic performance of the Abbott ARCHITECT HCV Ag assay for diagnosing active hepatitis C. The assay showed excellent sensitivity, specificity, and accuracy. However, its diagnostic utility may be limited in low prevalence settings, and further confirmation is recommended.

Summary: Despite individual variation in the clinical presentation of COVID-19, schizophrenia patients show a higher risk and severity of the disease compared to control counterparts. The genetic risk for schizophrenia, but not for bipolar disorder or depression, is associated with a higher risk of SARS-CoV-2 infection and COVID-19 severity, especially among women. However, the predictive accuracy is limited.

A Bayesian Multivariate Gametic Model in a Reciprocal Cross with Genomic Information: An Example with Two Iberian Varieties

Summary: INGA FOOD, S.A. conducted a crossbreeding program to produce hybrid pigs, and studies revealed differences in litter size between reciprocal crosses, suggesting the presence of genomic imprinting effects. A multivariate gametic model was introduced to estimate the gametic correlations between paternal and maternal effects from different genetic backgrounds. The results showed distinct differences in the posterior distribution of gametic correlations between the two populations, which may explain the performance outcomes in the reciprocal crosses.

Predictive plasma biomarkers of long-term increase in hepatic steatosis index after HCV eradication in HIV/HCV-coinfected patients

Summary: This study aimed to evaluate the association between plasma immune checkpoint proteins at baseline with hepatic steatosis index increase at the end of follow-up in HIV/HCV-coinfected patients. The results showed that higher levels of immune checkpoint proteins BTLA, CD137, CD80, GITR, LAG-3, and PD-L1 before HCV therapy were associated with a long-term increase in hepatic steatosis index after successful HCV therapy, suggesting a potential predictive role for early detection of progression towards steatosis in HIV/HCV-coinfected patients.

A multivariate gametic model for the analysis of purebred and crossbred data. An example between two populations of Iberian pigs

Summary: We investigated the relationship between HLA polymorphisms and susceptibility to SARS-CoV-2 infection and disease severity. Genotyping data from 9373 COVID-19-positive cases and 5943 population controls were analyzed. The study found associations between HLA-B*14:02 and HLA-C*08:02 alleles with a lower risk of COVID-19 infection, and HLA-A*11:01 and HLA-C*04:01 alleles with disease severity.
