Summary: In this study, the presence and subunit composition of protein complexes involved in energy conservation processes in anaerobic methanotrophic archaea were investigated using complexome profiling. All enzyme complexes and their subunit composition related to reverse methanogenesis were identified, providing a refined picture of the energy metabolism of these environmentally important microorganisms.

Ablation of mitochondrial DNA results in widespread remodeling of the mitochondrial complexome

Regulation of mitochondrial proteostasis by the proton gradient

Summary: Mitochondrial proteases, specifically the m-AAA protease AFG3L2, are regulated by the mitochondrial proton gradient and play a crucial role in reshaping the mitochondrial proteome in response to different energetic demands. TMBIM5, a Ca2+/H+ exchanger in the mitochondrial inner membrane, inhibits the activity of the m-AAA protease and ensures cell survival and respiration. However, persistent hyperpolarization triggers the degradation of TMBIM5 and activation of the m-AAA protease, which facilitates the proteolytic breakdown of respiratory complex I to limit ROS production in hyperpolarized mitochondria.

MRPS36 provides a structural link in the eukaryotic 2-oxoglutarate dehydrogenase complex

Summary: The tricarboxylic acid cycle is a central pathway for energy production in eukaryotic cells and plays a key role in aerobic respiration across all life kingdoms. The 2-oxoglutarate dehydrogenase complex (OGDHC) is a crucial enzyme in this cycle, generating NADH by oxidatively decarboxylating 2-oxoglutarate to succinyl-CoA. We provide evidence that MRPS36 is an important component of eukaryotic OGDHC, supported by cross-linking mass spectrometry data and phylogenetic analyses. We propose that MRPS36 evolved as an E3 adaptor protein, functionally replacing the peripheral subunit-binding domain (PSBD) in eukaryotic E2o.

Summary: The Cytb subunit of yeast mitochondrial complex bc is encoded in the mitochondrial genome and has eight transmembrane helices containing two hemes b. Cbp3 and Cbp6 assist in Cytb synthesis, while Cbp4 induces Cytb hemylation. Lack of Qcr7 reduces Cytb synthesis through an assembly-feedback mechanism involving Cbp3/Cbp6. Deletion of the Cytb C-region does not affect Cytb synthesis but disrupts the assembly-feedback regulation, resulting in non-respiratory mutants.
