Synthesis of Axially Chiral N-Arylindoles via Atroposelective Cyclization of Ynamides Catalyzed by Chiral Bronsted Acids

Summary: In recent years, there has been significant attention on the asymmetric catalysis of ynamides, with most reactions focused on central chirality. However, the synthesis of axially chiral compounds from ynamides has been limited to noble-metal catalysis. This study presents a metal-free protocol for the construction of axially chiral N-heterocycles from ynamides using chiral Bronsted acid-catalyzed 5-endo-dig cyclization. The method allows for the practical and atom-economical synthesis of valuable N-arylindoles with excellent enantioselectivities, and the resulting axially chiral N-arylindole skeletons can be employed in asymmetric catalysis.

Enantioselective Desymmetrizing Hydroalkoxylation of 1,4-and 1,8-Diynes Enabled by Chiral Bronsted Acid Catalysis

Summary: Herein, a chiral Bronsted acid-catalyzed desymmetrization reaction of diynes is reported, providing a facile access to enantioenriched 1,3-diaminopropanol derivatives and γ-butyrolactones with high enantioselectivities. These products can be used as precursors for the synthesis of versatile N- and O-heterocycles, which are important structural cores of bioactive molecules. Control experiments and theoretical calculations are used to confirm the mechanism and elucidate the origin of enantioinduction.

Summary: This study investigates the highly enantioselective intramolecular hydroamination reaction and uncovers its uncommon reaction mechanism. The reactivity of this reaction is primarily determined by the substituents on the internal carbon of the alkene, as well as the substituents on the terminal alkene carbons. The lower acidity of chiral phosphoric acids compared to chiral NTPAs results in a slower reaction.

Enantioselective Seleno-Michael Addition Reactions Catalyzed by a Chiral Bifunctional N-Heterocyclic Carbene with Noncovalent Activation

Summary: The Michael reaction is an important method for preparing functional molecules with a β-stereogenic center. However, the enantioselective seleno-Michael addition is still poorly explored. In this study, a highly enantioselective Michael addition reaction of alkyl selenols to enones was reported, introducing a Se atom asymmetrically to an electron-deficient double bond. A chiral bifunctional N-heterocyclic carbene (NHC)/thiourea catalyst was developed and showed remarkable selectivity in delivering chiral β-seleno ketones.

Insights into the Chiral Phosphoric Acid-Catalyzed Dynamic Kinetic Asymmetric Hydroamination of Racemic Allenes: An Allyl Carbocation/Phosphate Pair Mechanism

Summary: Computational studies of CPA-catalyzed DyKAH reactions reveal a catalytic asymmetric model involving pi-allylic carbocationic intermediates, with high E/Z selectivity achieved through intermolecular proton transfer. Noncovalent interactions within the electrostatic environment created by the phosphoric acid are key to explaining the enantioselectivity, while the chiral SPINOL scaffolds control the signs of enantioselectivity through substrate binding pocket configurations. Overall, understanding these mechanisms is crucial for guiding DyKAH reactions.

Selective Synthesis of C1-Symmetric BINOL-phosphates and P-chiral Phosphoramides Using Directed ortho-Lithiation

Summary: Directed ortho-lithiation (DoL) is an effective method for the ortho-substitution of BINOL-phosphoric acid and BINOL-N-triflylphosphoramide, allowing for rapid assembly of mono- or disubstituted BINOL-P-acids, including unsymmetrical disubstitution. Most significantly, DoL has proven to be highly effective in the diastereoselective desymmetrization of pseudo-C-2-symmetric BINOL-N-triflylphosphoramide, resulting in a chiral P-group.

Design and catalytic atroposelective synthesis of axially chiral isochromenone-indoles

Summary: The catalytic atroposelective synthesis of axially chiral isochromenone-indoles has been achieved using homophthalic anhydride-based indole derivatives as a new platform molecule for dynamic kinetic resolution. A wide range of axially chiral isochromenone-indoles were synthesized in high yields and excellent enantioselectivities using chiral quaternary ammonium salt as a phase-transfer catalyst in the catalytic asymmetric sulfonylation reaction.

Towards Axially Chiral Pyrazole-Based Phosphorus Scaffolds by Dipeptide-Phosphonium Salt Catalysis

Summary: In this article, a general and modular protocol for the synthesis of axially chiral arylpyrazole scaffolds containing a phosphorus unit is described. The reaction is catalyzed by a dipeptide phosphonium salt and involves an oxidative central-to-axial chirality conversion. The reaction demonstrates excellent yields and enantioselectivities, a broad substrate scope, and a low catalyst loading, leading to the formation of axially chiral phosphine compounds.

Summary: This work describes a chiral aldehyde/palladium catalysis method for asymmetric α-allylation of NH2-unprotected amino acid esters with 1,3-disubstituted allyl acetates. Using different chiral phosphine ligands, both anti- and syn-selective allylation reactions are achieved enantioselectively. A series of α,α-disubstituted amino acid esters with two adjacent chiral centers are produced in moderate-to-excellent yields, diastereoselectivities, and enantioselectivities.

Bronsted Acid Catalyzed Dearomatization by Intramolecular Hydroalkoxylation/Claisen Rearrangement: Diastereo- and Enantioselective Synthesis of Spirolactams

Summary: A novel Brønsted acid catalyzed intramolecular hydroalkoxylation/Claisen rearrangement is described for the synthesis of valuable spirolactams from readily available ynamides with good yields, diastereoselectivities, and broad substrate scope. An unexpected dearomatization of nonactivated arenes and heteroaromatic compounds occurs in this reaction. Furthermore, an asymmetric version of this reaction was achieved through efficient kinetic resolution by chiral phosphoric acid catalysis, with theoretical calculations showing a preference for the [3,3]-rearrangement over the [1,3]-rearrangement.

Design, Synthesis and Application of C2-Symmetric Cycloglycerodiphosphate Catalysts

Summary: Chiral phosphoric acids (CPAs) and their salts have been widely used as privileged catalysts in asymmetric transformations. Previous efficient CPAs have relied on C-2 symmetry and axial chirality, with the only exception being TADDOL derivatives. In this study, we have designed and synthesized C-2 symmetric cycloglycerophosphates (cGPAs), which were applied in the addition of trimethylsilyl cyanide (TMSCN) to aldehydes and ketones. The corresponding cyanohydrins were obtained with yields between 77% and 99% and enantiomeric excesses between 84.2% and 99.5%, in 27 examples. A DFT computational model was also used to rationalize the reaction mechanism and stereochemical outcome.
