Comparison of Four Different Preparation Methods for Making Injectable Microgels for Tissue Engineering and Cell Therapy

Summary: This study compares four different bioprocessing techniques for producing hydrogel microcarriers, aiming to achieve effective cell encapsulation while maintaining high cell viability and small particle size for injectability. The results show that the reverse thermal gelation technique produces cell-laden microgels with minimal specialized equipment and bioprocessing steps. The average microgel size is significantly smaller and more uniform when produced without cells.

Summary: This study further investigated the delivery strategy of using PF microspheres as a carrier for AON therapy in DMD. The results showed that the use of PF microspheres as a carrier improved the efficacy of AON therapy, restoring Dystrophin expression in mdx mice muscles and improving tissue histopathology. Intra-femoral injection of the microspheres demonstrated better results compared to intramuscular injection.

Modulus-dependent effects on neurogenic, myogenic, and chondrogenic differentiation of human mesenchymal stem cells in three-dimensional hydrogel cultures

Summary: This study evaluates how mechanical properties of a scaffold can alter the phenotype and genotype of human mesenchymal stromal cells (hMSCs), aiming to enhance their differentiation potential along the myogenic, neurogenic, or chondrogenic lineages. The shear storage modulus of a hydrogel scaffold was controlled by adjusting the amount of cross-linker, and it was found that materials with lower storage moduli promoted neurogenic differentiation, while those with intermediate moduli favored myogenic differentiation, and higher moduli favored chondrogenic differentiation.
