Summary: Untargeted separation of isomeric and isobaric species in mass spectrometry imaging is challenging. This study implemented nano-DESI MSI on a TIMS mass spectrometer and achieved high ion mobility resolution, successfully separating lipid isomers and isobars and revealing their distinct localizations in tissue samples. The results demonstrate the power of high-mobility-resolution IMS for analyzing complex biomolecular mixtures in MSI experiments.

Summary: The development and validation of desorption/ionization mass spectrometric assays for drug quantification in tissue sections is crucial for their widespread application in (clinical) pharmacology. Advances in desorption electrospray ionization have demonstrated its usefulness for developing targeted quantification methods that meet validation requirements. Factors such as desorption spot morphology, analytical time, and sample surface need to be considered for successful method development. This study highlights the importance of considering desorption kinetics during DESI analyses, which can help reduce analytical time, verify solvent-based drug extraction methods, and predict feasibility of imaging assays. These findings provide valuable guidance for future development of validated DESI-profiling and imaging methods.

Summary: In this paper, a new and easy to assemble dielectric barrier discharge plasma ionization source is presented, which is based on printed circuit boards with two parallel isolated electrodes for generating plasma inside an inert fused silica capillary. The plasma source is coupled to an ion mobility spectrometer for demonstration. Different ionization pathways can be achieved by whether the analytes pass through the plasma or bypass it before entering the reaction region of the ion mobility spectrometer. The plasma source has been designed using electric field simulations and can reach detection limits in the mid pptv range.

Summary: We assessed the complementarity and complexity of data detected in mammalian lipidome mass spectrometry imaging (MSI) using MALDI and nano-DESI. Our results showed that while MALDI had greater molecular coverage and dynamic range capabilities, nano-DESI provided superior mass error. Both modes demonstrated comparable degrees of lipidome complexity. We also evaluated the need for ultrahigh mass resolving power for certain mass splits in advanced instrumentation.

This review discusses the application of DESI-MSI and MALDI-MSI in evaluating exogenous and endogenous molecules in biological samples, providing technical insights and recent research findings.

Summary: Nanospray desorption electrospray ionization (nano-DESI) was used to analyze the metabolite fingerprints on Hypericum leaves. The intensity changes of key metabolites over time were observed through full mass scan experiments and monitored using selected ion monitoring and pseudo-selected reaction monitoring experiments. The correlation between the time-dependent changes in the metabolite fingerprints and depth-wise metabolite distribution was examined by removing the surface layers of the leaves multiple times.

Summary: This study investigated the performance of an electrospray ionization ion mobility spectrometer by varying the electric field strength in the desolvation region, introducing a new tristate ion shutter to enhance sensitivity for ions with higher molecular mass and lower ion mobility. The optimal ratio of electric field strengths in the drift and desolvation region was found to be affected by the electric field in the tristate ion shutter. The solvent flow rate also needs to be considered when setting the field strengths in the desolvation region to maintain proper electrospray function.

By using different combinations of fluorinated alcohols and alkylamines, it is possible to selectively reduce the charge states of oligonucleotides.
