Summary: Idiopathic pulmonary fibrosis (IPF) is a chronic disease characterized by scar tissue formation in the lungs. In this study, the researchers found that IPF patient-derived pulmonary ECM can induce fibrotic response in normal human lung fibroblasts. They identified GDF15 as one of the significantly upregulated proteins in the diseased ECM, and demonstrated its role in fibroblast activation and differentiation in promoting lung fibrosis.

Summary: In this study, the anti-fibrotic effects of derrone on TGF-beta 1-stimulated MRC-5 lung fibroblast cells and bleomycin-induced lung fibrosis were investigated. It was found that high concentrations of derrone increased cytotoxicity in MRC-5 cells, but low concentrations did not induce significant cell death. Derrone reduced the expression of TGF-beta 1, fibronectin, elastin, and collagen1 alpha 1 in TGF-beta 1-stimulated MRC-5 cells, and also attenuated fibrotic histopathological changes in bleomycin-treated mice.

Summary: This study investigated the effects of a selective CB1R agonist, ACPA, on pulmonary fibrosis induced by bleomycin (BLM). The results showed that ACPA treatment significantly improved survival rate and alleviated pulmonary fibrosis in BLM-treated mice. ACPA inhibited the expressions of extracellular matrix markers and diminished fibroblast migration triggered by TGF-beta. Furthermore, ACPA reduced the activation of TGF-beta-Smad2/3 signaling and decreased the expressions of downstream effector proteins in TGF-beta-challenged primary lung fibroblasts. These findings suggest that ACPA exhibits antifibrotic efficacy by targeting CB1R and inhibiting TGF-beta-Smad2/3 signaling.

Summary: The study demonstrates that fibroblast growth factor-21 (FGF-21) has significant preventive and therapeutic effects on pulmonary fibrosis, reducing the expression levels of related genes and proteins, inhibiting the aggregation of inflammatory cells and collagen deposition, and suppressing the expression of transforming growth factor-beta and alpha-smooth muscle actin in the lung. Therefore, FGF-21 could be a potential drug for the prevention and treatment of pulmonary fibrosis.
