Summary: Antibody-drug conjugates (ADCs) have made significant advancements in clinical cancer treatments, but are plagued by issues like low drug/antibody ratio and the need for large amounts of antibody. Researchers have developed an intelligent cell-selective nanotoxin with high specificity and cytotoxicity towards various cancer cells, showing significant inhibition effects. This nanotoxin appears to be a better platform than ADCs for targeted cancer therapy.

Summary: The development of a bioadhesive nanoparticle (BNP) drug delivery system significantly increased the therapeutic efficacy of intratumoral drug delivery by enhancing binding to SCC tumor cell surfaces and matrix proteins. In a mouse model, BNP-CPT markedly reduced tumor burden and showed signs of histologic cure in a portion of the lesions.

Summary: In this study, self-assembling genetically engineered polymeric nanoparticles formed by ELRs successfully inhibited the metabolic activity and cell viability of pancreatic cancer cells by blocking Akt protein phosphorylation and triggering apoptosis. These nanoparticles showed long circulating time and minimum toxicity in vivo, demonstrating potential for application in pancreatic cancer treatment.

Summary: Pancreatic cancer is the fourth leading cause of death worldwide, with low survival rates. The difficulties in early detection and the unique characteristics of pancreatic cancer contribute to ineffective therapies and widespread metastasis. Novel approaches, such as combining nanotechnology with cancer targeting, have shown potential in improving treatment efficacy and reducing cytotoxic effects.

Summary: Based on the immune escape and homologous adhesion ability of cancer cells, a biomimetic drug delivery system was developed by coating cancer cell membrane (CCM) on poly lactic acid-glycolic acid (PLGA) nanoparticles. The CCM-coated PLGA nanoparticles (CCMNPs) showed superior cellular affinity and had the abilities of immune escape and homologous targeting, which was confirmed by cellular uptake and in vivo distribution tests. Furthermore, CCMNPs achieved improved therapeutic effect of tumor and accurately targeted therapy, as evidenced by the higher tumor inhibition rate compared to PLGANPs and PTX injection.

Summary: This review discusses the serious drawbacks of conventional treatment for pancreatic ductal adenocarcinoma (PDAC) and the development of small interfering RNA (siRNA)-based therapeutics. Recent advances in drug delivery systems offer hope for improving treatment and developing nano-siRNA drugs. However, there are still many questions to address before clinical translation, such as toxicity, immunostimulation, and nano-bio interactions.

Summary: This study develops a simple and versatile biological drug delivery system (bDDS) based on human red blood cell-derived vesicles (RDVs), which shows excellent biocompatibility for cancer chemotherapy. The bDDS, named Dox-gluRDVs, effectively suppress multidrug resistance in uterine sarcoma by inducing cell death through a lysosomal-mitochondrial axis-dependent mechanism.

Summary: The limited availability of anticancer drugs and drug resistance have made monotherapy-based cancer therapy challenging. Combination therapy using currently available drugs is a potential alternative, but the barrier between in vitro screening and in vivo delivery hinders its application. This study proposes a precise drug delivery strategy using engineered nanomedicine to overcome this limitation and facilitate individualized combination therapy.
