γδ T Cells and Tumor Microenvironment: From Immunosurveillance to Tumor Evasion

gamma delta T cells possess cytotoxic antitumor activity mediated by production of proinflammatory cytokines, direct cytotoxic activity, and regulation of the biological functions of other cell types. Hence, these features have prompted the development of therapeutic strategies in which gamma delta T cells agonists or ex vivo-expanded gamma delta T cells are administered to tumor patients. Several studies have shown that gamma delta T cells are an important component of tumor-infiltrating lymphocytes in patients affected by different types of cancer and a recent analysis of similar to 18,000 transcriptomes from 39 human tumors identified tumor-infiltrating.d T cells as the most significant favorable cancer-wide prognostic signature. However, the complex and intricate interactions between tumor cells, tumor microenvironment (TME), and tumor-infiltrating immune cells results in a balance between tumor-promoting and tumor-controlling effects, and gamma delta T cells functions are often diverted or impaired by immunosuppressive signals originating from the TME. This review focuses on the dangerous liason between gamma delta T cells and tumoral microenvironment and raises the possibility that strategies capable to reduce the immunosuppressive environment and increase the cytotoxic ability of gamma delta T cells may be the key factor to improve their utilization in tumor immunotherapy.

Summary: This review comprehensively analyzes the effects of SARS-CoV-2 infection on Unconventional T cells and innate lymphoid cells (ILCs). The dysregulation of adaptive immune systems in COVID-19 affected patients has been observed, but there are still many unanswered questions regarding the behavior of Unconventional cells and ILCs during infection. This analysis highlights the differences in studies on patient cohorts and explores potential diagnostic factors and targeted therapies that have been overlooked before. Monitoring the frequencies and functions of Unconventional T cells and ILCs in longitudinal studies may provide useful prognostic biomarkers and future therapeutic strategies for SARS-CoV-2-infected individuals.

A Mathematical Model of In Vitro Cellular Uptake of Zoledronic Acid and Isopentenyl Pyrophosphate Accumulation

Summary: The mevalonate pathway is a promising target for various research areas, and blocking this pathway with drugs like zoledronic acid can inhibit cancer cell growth. This study proposes two mathematical models to describe the kinetics of isopentenyl pyrophosphate (IPP) accumulation and its relationship with zoledronic acid treatment. The models fit the experimental data well and provide insights into the pharmacodynamics of IPP.

Autologous conditioned serum for chronic pain in patients with osteoarthritis: A feasibility observational study

Summary: This pilot observational study evaluated the feasibility of conditional autologous serum (ACS) injections in patients with severe chronic pain and grade I-III osteoarthritis. The study also assessed the cytokine content of the ACS. The results showed a significant reduction in pain intensity after 6 months of ACS injections, suggesting that ACS may be a potential treatment option for patients with refractory osteoarthritis.

Summary: In recent years, research has focused on implementing modern treatment approaches to improve patient survival in colorectal cancer. Gamma delta T cells are being explored as a new and promising candidate for treating various types of cancer due to their potent killing activity and ability to recognize tumor antigens independently of HLA molecules. This article specifically focuses on the role of gamma delta T cells in antitumor immunity, particularly in colorectal cancer, and highlights small-scale clinical trials in which in vivo activation or adoptive transfer of ex vivo expanded gamma delta T cells were employed for the treatment of colorectal cancer. Possible combinatorial approaches to treat colon cancer are also suggested.

Ferritin Metabolism Reflects Multiple Myeloma Microenvironment and Predicts Patient Outcome

Summary: Multiple myeloma (MM) is a hematologic malignancy characterized by a pro-inflammatory and immunosuppressive microenvironment. Ferritin, released by pro-inflammatory cells, contributes to ROS production and cellular damage. The study found that high ferritin levels were associated with worse prognosis in MM patients and correlated with systemic inflammation markers and specific bone marrow cell microenvironment. Bioinformatic analysis also revealed that ferritin biosynthesis gene expression signature was associated with poor outcome and specific immune cell profiles. These findings suggest that ferritin could be a potential target for improving MM patient outcome.

An analysis of the immunomodulatory properties of human spheroids from adipose-derived stem cells

Summary: This study analysed the immunomodulatory properties of spheroids from adipose stem cells (SASCs) and compared them to adipose stem cells (ADSCs). SASCs showed stronger immunomodulatory abilities in heterologous experimental settings compared to ADSCs. SASCs also demonstrated greater anti-inflammatory properties.

Lemon-derived nanovesicles achieve antioxidant and anti-inflammatory effects activating the AhR/Nrf2 signaling pathway

Summary: In recent years, extracellular vesicles (EVs) from various plant sources have attracted the attention of the scientific community due to their interesting biological properties. This study isolated and characterized nano-vesicles from lemon juice (LNVs) and investigated their antioxidant effects. LNV pre-treatment reduced ROS levels in fibroblasts stimulated with H2O2 and UVB, and this reduction was associated with the activation of the AhR/Nrf2 signaling pathway. In zebrafish embryos, LNVs also exhibited antioxidant effects by reducing ROS levels and neutrophil migration when stimulated with LPS.

Monoclonal Gammopathies and the Bone Marrow Microenvironment: From Bench to Bedside and Then Back Again

Summary: Multiple myeloma (MM) is an incurable hematologic malignancy with a multistep evolutionary pathway, involving monoclonal gammopathy of undetermined significance (MGUS), smoldering MM (sMM), and symptomatic MM. Genomic alterations are present at the MGUS phase, but their role in disease evolution is questionable. Changes in the bone marrow microenvironment (TME) play a key role in MM evolution, driving cancer progression and clonal plasma cell behavior. Microenvironment-oriented drugs have significantly improved MM patients' survival. This review discusses the role of the TME in MM evolution and potential therapeutic targets/opportunities.
