Co-protoporphyrin IX and Sn-protoporphyrin IX inactivate Zika, Chikungunya and other arboviruses by targeting the viral envelope

The global situation of diseases transmitted by arthropod-borne viruses such as Dengue (DENV), Yellow Fever (YFV), Chikungunya (CHIKV) and Zika (ZIKV) viruses is alarming and treatment of human infection by these arboviruses faces several challenges. The discovery of broad-spectrum antiviral molecules, able to inactivate different groups of viruses, is an interesting approach. The viral envelope is a common structure among arboviruses, being a potential target for antivirals. Porphyrins are amphipathic molecules able to interact with membranes and absorb light, being widely used in photodynamic therapy. Previously, we showed that heme, Co-protoporphyrin IX (CoPPIX) and Sn-protoporphyrin IX (SnPPIX) directly inactivate DENV and YFV infectious particles. Here we demonstrate that the antiviral activity of these porphyrins can be broadened to CHIKV, ZIKV, Mayaro virus, Sindbis virus and Vesicular Stomatitis virus. Porphyrin treatment causes viral envelope protein loss, affecting viral morphology, adsorption and entry into target cells. Also, light-stimulation enhanced the SnPPIX activity against all tested arboviruses. In summary, CoPPIX and SnPPIX were shown to be efficient broad-spectrum compounds to inactivate medically and veterinary important viruses.

Summary: Two platforms based on reduced graphene oxide functionalized with polymers and loaded with amphotericin B were fabricated and tested against Leishmania amazonensis. Among them, rGO-PEI-AmB showed more pronounced activity against the parasite and better performance under near-infrared light irradiation, making it a promising candidate for the topical treatment of cutaneous leishmaniasis.

Searching for drug leads targeted to the hydrophobic cleft of dengue virus capsid protein

Summary: A study synthesised and screened 18 aromatic derivatives of guanylhydrazones and oximes aromatic for their ability to bind to dengue virus capsid protein. Results showed that five compounds were able to bind to a specific region of the capsid protein, inhibiting the virus infectivity.

Inhibiting ROS generation by targeting mitochondrial or NOX pathways can reduce virus replication, cell death, and inflammation, presenting potential therapeutic targets for preventing dengue pathogenesis.

Summary: This study presents the fabrication of two light-responsive systems based on functionalized reduced graphene oxide (rGO) loaded with the antileishmanial drug AmB. The systems were characterized using various methods and the drug release and photothermal effects were investigated. Optimization of the drug and polymer structures was performed and the antiproliferative activity against Leishmania amazonensis promastigotes was evaluated.

Summary: Cognitive dysfunction in post-COVID-19 syndrome may be caused by the release of SARS-CoV-2 Spike protein, which can lead to neuroinflammation, synapse elimination, and memory dysfunction mediated by Toll-like receptor 4 (TLR4) signaling. Infusing Spike protein into the brains of mice reproduced cognitive impairment similar to post-COVID-19 syndrome. In a group of 86 mild COVID-19 patients, a specific TLR4 genotype was associated with poor cognitive outcome. These findings highlight TLR4 as a key target for investigating long-term cognitive dysfunction after COVID-19 infection.

Summary: In this study, different outcomes were observed in mice infected with Zika virus (ZIKV) based on age and sex: neonatally infected animals showed an increase in prepulse inhibition (PPI) and delayed startle latency, while adult-infected male mice had lower startle amplitude and both sexes showed PPI impairment 14 days after infection. These findings contribute to a better understanding of the functional impacts of ZIKV on the nervous system and provide insight into the behavioral and cognitive alterations caused by the virus. The study supports the use of startle reflex testing in ZIKV-exposed patients, especially infants, for early detection of functional neuromotor damage and early intervention.

Accuracy of a raw saliva-based COVID-19 RT-LAMP diagnostic assay

Summary: The SARS-CoV-2 pandemic called for rapid diagnosis of COVID-19 cases to prevent disease transmission. This study validates the use of RT-LAMP in saliva samples and demonstrates high accuracy in purified RNA-saliva samples. Despite mass vaccination, the presence of new strains and slow progress in vaccinations contribute to the spread of COVID-19. This study shows that tracking new COVID-19 cases in a large population is feasible using raw saliva samples in the RT-LAMP assay, which provides accurate results and a less invasive testing option.

In Silico Physicochemical Characterization of Fusion Proteins from Emerging Amazonian Arboviruses

Summary: The Mayaro (MAYV), Saint Louis encephalitis (SLEV), and Oropouche (OROV) viruses are neglected arboviruses that cause febrile illnesses in humans in the Amazon region. This study conducted an in silico analysis to characterize the physicochemical properties of their fusion proteins. The results showed that the MAYV E1 and SLEV E proteins share more similarities in physicochemical characteristics compared to the OROV Gc protein.

Summary: This study aimed to evaluate the correlation between serum antibody seroconversion and neutralizing antibody titers in individuals who received a complete initial vaccination schedule. The results showed that although IgG titers were high in all individuals after vaccination, the amounts of neutralizing antibodies varied among the groups. Serum neutralizing antibody concentrations were more comparable to IgA levels, suggesting that these antibodies have a major neutralizing capacity against SARS-CoV-2.
