Summary: This study demonstrates that temporary exposure to PM2.5 induces lung epithelial cell apoptosis through promoting cytokines expression and inflammatory factors secretion. The cytotoxicity of PM2.5 can be alleviated by tussilagone (TSL), which restores the Hif-1 alpha protein level and decreases NF-kappa B activation. TSL treatment effectively relieves the damage caused by PM2.5 and decreases the expression of inflammatory cytokines.

Summary: In this study, we found that the use of UPR inducer Tunicamycin can protect against injury caused by lipopolysaccharide. By modulating actomyosin remodeling and enhancing endothelial integrity, TM supports vascular barrier function. Our findings suggest that UPR activation may provide novel therapeutic opportunities for diseases related to endothelial dysregulation.

Summary: This study discovered a natural compound, licochalcone A, with anti-inflammatory activity. It was found to directly bind to the protein MD2, which regulates the LPS inflammatory signaling pathway, and blocked both TRIF- and MYD88-dependent pathways. In vivo experiments showed that licochalcone A treatment alleviated acute lung injury (ALI) in mice by reducing immunocyte infiltration and suppressing TLR4 pathway activation and inflammatory cytokine production.

Summary: This study investigated the anti-inflammatory mechanism of Schisandrin B (Sch B) in macrophages and an animal model of acute lung injury (ALI) and found that Sch B directly binds to MyD88 to regulate inflammatory responses and inhibit the expression of inflammatory factors. The study suggests that Sch B has therapeutic potential for ALI and other MyD88-dependent inflammatory diseases.

Summary: Local delivery of vitamin D to the lungs efficiently reduced inflammatory cells induced by LPS in bronchoalveolar lavage and slightly attenuated an increase in CXCL5. In cases of severe vitamin D deficiency, despite not significantly minimizing cellular inflammation, local vitamin D prevented epithelial barrier leakage and damage in the lung. Additional research is needed to determine the long-term beneficial effects of local vitamin D delivery on lung inflammation.

Summary: This study found that after exposure to paraquat, oxygen free radicals are generated, causing lipid peroxidation of tissue and organ cell membranes. This leads to multiple organ dysfunction, with acute lung injury being the most common and severe. The specific mechanism of acute lung injury caused by paraquat is still unclear. The study suggests that HIF-1 alpha may promote oxidative stress and inflammatory response in alveolar epithelial cells by regulating the expression of Rac2, thereby participating in paraquat-induced acute lung injury.

Summary: Mesenchymal stem cells (MSCs) therapy attenuates LPS-induced acute lung injury and Pseudomonas aeruginosa (PA)-induced acute pneumonia, but the underlying molecular mechanisms are unclear. This study shows that PA-derived outer membrane vesicles (OMVs) induce mouse primary adipose tissue-derived mesenchymal stem cells (ASCs) to upregulate cyclic GMP-AMP synthase (cGAS) and express interleukin (IL)-7. Loss of cGAS-interferon (IFN)-beta axis abolishes the protective function of ASCs against PA-induced acute pneumonia. OMVs-delivered PA dsDNA activates cGAS-STING signaling pathway and increases IL-7 production via IFN-beta signaling in ASCs, which further amplifies IL-7 expression in lung epithelial cells.
