Summary: This study suggests that BDNF deficient mice are more vulnerable to depressive-like behaviors following peripheral immune challenge, displaying an exaggerated neuroinflammatory response. While behavioral despair and anxiety-like behavior did not differ between genotypes, BDNF+/- mice showed significantly more pronounced anhedonia-like behavior after LPS injection compared to BDNF+/+ mice.

Summary: We found that insulin treatment significantly improved LPS-induced cognitive decline, neuronal injuries, and blood-brain barrier disruption. Insulin treatment inhibited the activation of microglia and astrocytes, and the release of TNF-α and IL-13 in the hippocampus. Furthermore, insulin treatment inhibited LPS-induced ferroptosis in the hippocampus by decreasing iron accumulation levels, regulating ferroptosis-related proteins, and inhibiting oxidative stress injuries and lipid peroxidation.

Therefore, GP-14 could be a promising lead compound for treating depression.
