Fast Profiling of Natural Pigments in Different Spirulina (Arthrospira platensis) Dietary Supplements by DI-FT-ICR and Evaluation of their Antioxidant Potential by Pre-Column DPPH-UHPLC Assay

Arthrospira platensis, better known as Spirulina, is one of the most important microalgae species. This cyanobacterium possesses a rich metabolite pattern, including high amounts of natural pigments. In this study, we applied a combined strategy based on Fourier Transform Ion Cyclotron Resonance Mass Spectrometry (FT-ICR-MS) and Ultra High-Performance Liquid Chromatography (UHPLC) for the qualitative/quantitative characterization of Spirulina pigments in three different commercial dietary supplements. FT-ICR was employed to elucidate the qualitative profile of Spirulina pigments, in both direct infusion mode (DIMS) and coupled to UHPLC. DIMS showed to be a very fast (4 min) and accurate (mass accuracy 0.01 ppm) tool. 51 pigments were tentatively identified. The profile revealed different classes, such as carotenes, xanthophylls and chlorophylls. Moreover, the antioxidant evaluation of the major compounds was assessed by pre-column reaction with the DPPH radical followed by fast UHPLC-PDA separation, highlighting the contribution of single analytes to the antioxidant potential of the entire pigment fraction. -carotene, diadinoxanthin and diatoxanthin showed the highest scavenging activity. The method took 40 min per sample, comprising reaction. This strategy could represent a valid tool for the fast and comprehensive characterization of Spirulina pigments in dietary supplements, as well as in other microalgae-based products.

Application of Calcium Alginate Hydrogels in Semisolid Extrusion 3D Printed for the Production of Easy-to-Swallow Tablets

Summary: To fully utilize the potential of 3D printing, particularly in the case of semisolid extrusion (SSE), the technological challenges need to be addressed. This study focuses on investigating the physicochemical properties of natural polymers, specifically alginate hydrogel, for 3D printing tablets that are easy to swallow. By adding sorbitol as a plasticizer, the aim is to improve the swallowability of the tablets. Different concentrations of sorbitol solution are evaluated to determine the best composition in terms of printing and technological properties. The results highlight a predictable relationship between the amount of sorbitol loaded and the residual mass of the final platforms.

In Silico Identification and In Vitro Evaluation of New ABCG2 Transporter Inhibitors as Potential Anticancer Agents

Summary: Different mechanisms contribute to cancer multidrug resistance, including increased drug efflux, enhanced cellular repair mechanisms, and alterations of drug metabolism or targets. ABCG2, a transporter protein, promotes drug efflux and causes chemotherapeutic resistance in malignant cells. Through a multidisciplinary approach, a new selective inhibitor (8) of ABCG2 has been identified, which effectively improves the cytotoxicity of mitoxantrone in hepatocellular carcinoma and transfected breast cancer cell lines. This inhibitor also demonstrates its efficacy in reversing multidrug resistance with the overexpression of the ABCG2 pump.

ER stress, caused by disturbances in ER homeostasis, triggers unfolded protein response (UPR) to counteract the stress. Calcium levels are crucial in maintaining ER homeostasis and regulating cell fate, and dysregulation of calcium signaling is associated with diseases like neurodegeneration, cancer, and metabolic disorders. Recent research suggests that TRP channels play a role in UPR-mediated ER stress and can influence cell survival or death, making them a potential therapeutic target.

3D printed macroporous scaffolds of PCL and inulin-g-P(D,L)LA for bone tissue engineering applications

Summary: In this study, a PCL/INU-PLA hybrid biomaterial was prepared, which showed improved surface properties, swelling ability, and in vitro biodegradation rate compared to PCL alone. The 3D printed scaffolds exhibited dimensional and structural parameters close to the digital model and showed favorable results in biocompatibility and osteogenic activity tests.

Summary: This study describes the rational development of soluble epoxide hydrolase (sEH) inhibitors for the treatment of acute pancreatitis (AP). The most promising compound, 28, was identified through in vitro screening and molecular modeling studies. Compound 28 demonstrated remarkable efficacy in reducing inflammatory damage in a mouse model of cerulein-induced AP. Metabololipidomic analysis further confirmed the molecular mechanism of sEH inhibition in the anti-AP activity of compound 28. Overall, compound 28 shows strong effectiveness as an sEH inhibitor with potential for pharmacological treatment of AP.

Summary: A series of tetrasubstituted pyrrole derivatives (TSPs) were synthesized based on a previous hypothesis and showed significant toxicity against human epithelial melanoma A375 cells. The most active compound, 10a, resulted as the most soluble among previous and present TSPs and exhibited apoptotic cell-death pathways activation. The molecular mechanism of action based on the ability of TSPs to mimic the hydrophobic side chains of protein motifs was suggested.

Rational Design of Novel Peptidomimetics against Influenza A Virus: Biological and Computational Studies

Summary: This study aimed to synthesize a new library of peptidomimetics active against the influenza virus and conducted a preliminary screening using biophysical assays. Through biological and computational studies on the most interesting compounds, a small peptide sequence with high activity was identified, providing a good starting point for the design of new peptidomimetics and small molecules.

Couroupita guianensis bark decoction: From Amazonian medicine to the UHPLC-HRMS chemical profile and its role in inflammation processes and re-epithelialization

Summary: This study compared the metabolite profile of Couroupita guianensis bark decoction produced by Amazonian shamans with that obtained under laboratory conditions and investigated their biological properties in wound healing and inflammation. The analysis revealed the presence of polyphenolic compounds and uncommon sulphated derivatives of ellagic acid in the decoction. A new sulphated molecule was identified as the potential active compound responsible for wound healing. The decoction also showed anti-inflammatory effects and potential therapeutic applications in skin diseases.
