Viral metagenomics, genetic and evolutionary characteristics of Crimean-Congo hemorrhagic fever orthonairovirus in humans, Kosovo

Crimean-Congo hemorrhagic fever orthonairovirus (CCHFV) is a tick-borne virus which causes severe disease in humans with fatality cases up to 30%. We investigated the genetic and evolutionary characteristics of CCHFV in Kosovo, in particular in humans and found that different virus variants of genotype V circulate, with Turkey as a possible origin for the progenitor of southern European CCHF outbreaks. Phylogenetic analyses also revealed a single introduction event and in situ evolution of CCHFV in this country. The viral metagenomics revealed a more abundant virome in the fatal CCHF cases and the presence of a novel tick-borne segmented RNA virus belonging to the recently discovered Jingmenvirus group which raises questions about the potential pathogenic effect of this novel virus on human and animal health.

Summary: The study found that Lassa virus RNA can be detected in various body fluids after recovery from acute Lassa fever, with the longest persistence in seminal fluid. The results showed that the persistence of viral RNA in seminal fluid was associated with higher viral load, severity of disease, and longer hospitalization during the acute phase of Lassa fever.

Summary: This study evaluated the use of a capture enzyme-linked immunosorbent assay (ELISA) for detecting LASV-reactive IgG antibodies in Mastomys rodents. The ELISA showed a high accuracy of 97.1% in known exposure samples and exhibited strong agreement with an immunofluorescence assay (IFA). It can be used as an alternative to IFA for determining LASV IgG serostatus in Mastomys rodents.

Summary: Highly pathogenic Arenaviruses, like the Lassa Virus (LASV), pose a serious public health threat in affected countries. Research and development of vaccines and therapeutics are urgently needed but hampered by the necessity to handle these pathogens under biosafety level 4 conditions. These containment restrictions make large-scale screens of antiviral compounds difficult. Therefore, the Mopeia virus (MOPV), closely related to LASV, is often used as an apathogenic surrogate virus.

This study highlights the potential link between environmental change and LASV emergence, emphasizing the urgent need for research on land management practices that reduce disease risks.
