Hydroxytyrosol: Bioavailability, toxicity, and clinical applications

Many beneficial properties have been attributed to the Mediterranean diet. Over the years, researchers have attempted to learn which foods and which food components are responsible for good health. One of these components is hydroxytyrosol, an important phenolic compound present in olive oil. Hydroxytyrosol is a molecule of high interest to the pharmaceutical industry due to its anti-inflammatory and antimicrobial qualities its role against cardiovascular diseases and metabolic syndrome and for its neuroprotection, antitumour, and chemo modulation effects. The interest in this molecule has led to wide research on its biological activities, its beneficial effects in humans and how to synthetize new molecules from hydroxytyrosol. This review describes the vast range of information about hydroxytyrosol, focusing on its involvement in biological mechanisms and modulation effects on different pathologies. This review also serves to highlight the role of hydroxytyrosol as a nutraceutical and as a potential therapeutic agent.

Involvement of redox signalling in tumour cell dormancy and metastasis

Summary: Decades of research have revealed the complexity of tumour cellular and molecular biology, with some progress made in treating primary tumours. However, less progress has been made in developing therapies against metastasis, which is the main cause of cancer-related deaths. Recent studies have shown that epigenetic and microenvironmental factors play a key role in tumour progression. Free radicals act as second messengers and modulate tumour cell signalling, leading to either cell survival or cell death depending on the context. Redox signalling is involved in controlling disseminated tumour cell fate and metastasis.

Summary: The risk of cancer increases with age, but the development of estrogen-receptor-positive (ER+) cancer types in women is more related to menopausal status than age. Obesity-induced inflammation, a general risk factor for cancer, shows differences in its association with ER+ cancer risk in pre- and postmenopausal women. This study demonstrates the different biological functions of estradiol (E2) and estrone (E1) in ER+ gynecologic cancers, highlighting the potential role of E1 and HSD17B E1-synthesizing enzymes in the development and progression of these diseases.

Emerging Role of Plant-Based Dietary Components in Post-Translational Modifications Associated with Colorectal Cancer

Summary: Colorectal cancer (CRC) is a common cancer with modifiable risk factors including diet, alcohol consumption, and smoking. Natural dietary components have shown chemopreventive activity in CRC through modulation of cellular processes. Post-translational modifications (PTMs) of proteins associated with CRC have gained interest as inappropriate modification is linked to carcinogenesis. This review collects and analyzes the main PTMs associated with CRC, explores the relationship between different proteins susceptible to inappropriate PTMs, and reviews the literature on the role of plant-based dietary compounds in modulating CRC-associated PTMs. The review suggests that plant-based dietary components can correct inappropriate PTMs and promote apoptosis in tumor cells.

Molecular Mechanisms of the Protective Effects of Olive Leaf Polyphenols against Alzheimer's Disease

Summary: Alzheimer's Disease (AD) is a major cause of global dementia cases, affecting around 50 million people worldwide. The leaves of olive trees contain bioactive compounds such as oleuropein (OLE) and hydroxytyrosol (HT), which have shown medicinal properties in fighting AD. Olive leaf (OL), OLE, and HT can reduce amyloid-beta and neurofibrillary tangles formation, and OL has exhibited high cholinergic inhibitory activity. These compounds may also decrease neuroinflammation and oxidative stress, potentially promoting autophagy and restoring loss of proteostasis in AD models. Therefore, olive phytochemicals hold promise as an adjuvant in AD treatment.

Summary: This study aimed to evaluate the potential positive effects of ubiquinol supplementation on muscle aggression, physical performance, and fatigue perception in non-elite athletes after high intensity circuit weight training. The results showed that ubiquinol supplementation improved muscle performance, reduced muscle damage markers, and had a protective effect on muscle fibers.

Summary: Since the COVID-19 pandemic is ongoing, exploring strategies to support the immune system during infection is necessary. An adequate diet can improve immune responses, and minerals, which have immune-modulatory and antioxidant activities, might be useful in fighting this illness. Although more research is needed, evidence from studies on respiratory diseases suggests the rationality of investigating the use of minerals during this pandemic.

Influence of Adipose Tissue on Early Metabolic Programming: Conditioning Factors and Early Screening

Summary: Background: Obesity and being overweight have become severe global health issues. Maternal dietary habits and other factors during pregnancy can have a significant impact on the prevention of obesity in children. A review of recent human studies aims to determine modifiable factors that influence adiposity and prevent specific disorders later in life. Conclusion: A healthy diet and lifestyle starting from pregnancy and throughout childhood, particularly in the first two years, can prevent the emergence of noncommunicable diseases and metabolic syndrome-related pathologies.

Can the phenolic compounds of Manuka honey chemosensitize colon cancer stem cells? A deep insight into the effect on chemoresistance and self-renewal

Summary: This study investigated the chemosensitizing effects of Manuka honey against 5-Fluorouracil in colonspheres enriched with cancer stem cells (CSCs). The results showed that combining Manuka honey with 5-Fluorouracil decreased the gene expression of ABC sub-family G member 2 and thymidylate synthase, which are associated with drug resistance and DNA synthesis respectively. Manuka honey also regulated the Wnt/β-catenin, Hedgehog and Notch pathways, reducing the self-renewal ability of CSCs. This preliminary study opens new possibilities for the combination of natural compounds and pharmaceuticals to enhance efficacy or reduce adverse effects.

Lycopene Supplementation for Patients Under Cancer Therapy: A Systematic Review and Meta-Analysis of Randomized Controlled Trials

Summary: The study investigates the effects of lycopene supplementation on cancer therapy outcomes and circulating lycopene concentration in patients. The results show that lycopene supplementation does not modify cancer hallmarks but increases circulating lycopene concentration in patients under cancer therapy, which could have a positive impact on potential clinical and molecular outcomes in cancer patients.

Summary: In this study, olive leaves from different geographical origins were analyzed for their phytochemical profile, antioxidant capacity, neuroprotective activity, and anti-inflammatory effects. The results showed that olive leaves from Spain and Italy had the highest antioxidant and neuroprotective activities, while those from Greece had the lowest. The content of oleoside methyl ester and p-hydroxybenzoic acid in the Spanish and Italian samples, respectively, were strongly associated with these biological activities.

The application of ice glazing containing D-sodium erythorbate combined with vacuum packaging to maintain the physicochemical quality and sweet/umami non-volatile flavor compounds of frozen stored large yellow croaker (Pseudosciaena crocea)

Summary: This study investigated the use of ice glazing containing D-sodium erythorbate (DSE) and vacuum packaging to maintain the quality of large yellow croaker during frozen storage. The results showed that vacuum packaging effectively inhibited ice crystal growth and minimized water loss, while the combination of vacuum packaging and 0.3% DSE-infused ice glazing maintained freshness indicators at low levels throughout the 300-day storage period. This combination was considered the most effective in preserving the quality of the fish.

Hypoglycemic effect and intestinal transport of phenolics-rich extract from digested mulberry leaves in Caco-2/insulin-resistant HepG2 co-culture model

Summary: This study investigated the transport and hypoglycemic effects of phenolics in mulberry leaves using an in vitro digestion model. The results showed that digested phenolics had higher absorbability and could inhibit glucose digestion and absorption. Phenolics also regulated glucose metabolism. Luteoforol and p-coumaric acid were found to be the primary phenolics strongly correlated with the hypoglycemic ability of mulberry leaves.

Summary: This study proposed an integrated and automated procedure for extracting, separating, and quantifying bioactive compounds from coffee co-products. By optimizing the extraction, separation, and analysis method, real-time analysis was achieved. The results suggest that coupling different techniques can efficiently extract, separate, and analyze phenolic compounds, providing an integrated method to produce high-added value ingredients for various applications.

Geographical origin discriminatory analysis of onions: Chemometrics methods applied to ICP-OES and ICP-MS analysis

Summary: Geographical origin plays a crucial role in determining the quality and safety of agricultural products. In this study, ICP analysis was used to determine the inorganic elemental content of onions and identify their geographical origin. Chemometric methods were applied to analyze the ICP results, and the accuracy of distinguishing between Korean and Chinese onions was found to be excellent. The findings suggest that this method can be beneficial for identifying agricultural products.
