Endoperoxide-8-aminoquinoline hybrids as dual-stage antimalarial agents with enhanced metabolic stability

Hybrid compounds may play a critical role in the context of the malaria eradication agenda, which will benefit from therapeutic tools active against the symptomatic erythrocytic stage of Plasmodium infection, and also capable of eliminating liver stage parasites, To address the need for efficient multistage antiplasmodial compounds, a small library of 1,2,4,5-tetraoxane-8-aminoquinoline hybrids, with the metabolically labile C-5 position of the 8-aminoquinoline moiety blocked with aryl groups, was synthesized and screened for antiplasmodial activity and metabolic stability. The hybrid compounds inhibited development of intra-erythrocytic forms of the multidrug-resistant Plasmodium falciparum W2 strain, with EC50 values in the nM range, and with low cytotoxicity against mammalian cells. The compounds also inhibited the development of P. berghei liver stage parasites, with the most potent compounds displaying EC50 values in the low mu M range. SAR analysis revealed that unbranched linkers between the endoperoxide and 8-aminoquinoline pharmacophores are most beneficial for dual antiplasmodial activity. Importantly, hybrids were significantly more potent than a 1:1 mixture of 8-aminoquinoline-tetraoxane, highlighting the superiority of the hybrid approach over the combination therapy. Furthermore, aryl substituents at C-5 of the 8-aminoquinoline moiety improve the compounds' metabolic stability when compared with their primaquine (i.e. C-5 unsubstituted) counterparts. Overall, this study reveals that blocking the quinoline C-5 position does not result in loss of dual-stage antimalarial activity, and that tetraoxane-8-aminoquinoline hybrids are an attractive approach to achieve elimination of exo- and intraerythrocytic parasites, thus with the potential to be used in malaria eradication campaigns. Synthesis and antiplasmodial activity of regioisomers and epimers of second-generation dual acting ivermectin hybrids

Summary: With its strong impact on vector-borne diseases and malaria, ivermectin has been proven to be a promising antimalarial therapeutic. By modifying the structure of ivermectin and combining it with existing antimalarial agents, novel molecular hybrids with enhanced activity against Plasmodium parasites have been developed. These compounds displayed structure-dependent antiplasmodial activity, outperforming ivermectin itself, suggesting the superiority of this hybridization strategy in designing new antimalarial agents.

Target Identification in Anti-Tuberculosis Drug Discovery

Summary: The human immunodeficiency virus (HIV) causes acquired immunodeficiency syndrome (AIDS). An increase in viral load leads to a decline in T lymphocytes, compromising the immune system. Tuberculosis (TB) is the most common opportunistic disease in HIV-positive patients. Treatment for HIV-TB coinfection is challenging due to drug interactions, toxicity, non-adherence, and resistance. Recent approaches involve using molecules that target multiple distinct sites to improve therapy. This review discusses the importance of multitarget strategies and the development of structural entities for simultaneous treatment of HIV-TB.

Block Copolymer Micelles Encapsulating Au(III) Bis(Dithiolene) Complexes as Promising Nanostructures with Antiplasmodial Activity

Summary: Block copolymer micelles (BCMs) were evaluated as drug delivery systems for gold(III) bis(dithiolene) complexes (AuS and AuSe) to treat malaria. The complexes showed strong antiplasmodial activity and low toxicity when encapsulated in BCMs. Encapsulated complexes exhibited controlled release and improved biocompatibility compared to the non-encapsulated counterparts. This study suggests that BCMs can be a promising alternative for conventional antimalarial treatments.

The system can be easily adapted for other pathogens and has the potential for early diagnosis of infectious diseases.

The effect of dosage on the protective efficacy of whole-sporozoite formulations for immunization against malaria

Summary: Immunization with Plasmodium sporozoites, especially whole-sporozoite (WSpz) formulations, can provide strong protection against malaria. However, a comparative analysis of different WSpz formulations showed that early liver arresting genetically attenuated parasites (EA-GAP) formulation failed to confer complete protection at any immunization dosage, suggesting a minimum threshold of liver development is required for effective immune responses. On the other hand, radiation-attenuated sporozoites (RAS), late arresting GAP (LA-GAP), and sporozoites administered under chemoprophylaxis (CPS) formulations yielded comparable and dosage-dependent protection. Immunological analyses suggested that EA-GAP immunization may negatively impact memory cell functionality and protective immunity.

A pro-death autophagy-based nanoplatform for enhancing antitumour efficacy with improved immune responses

Furthermore, the study discovers a new therapeutic direction for the respiration inhibitor 3-bromopyruvic acid (3BP) in cancer treatment. Overall, this study offers an opportunity to improve antitumor efficacy and boost immune responses.

A novel scaffold long-acting selective estrogen receptor antagonist and degrader with superior preclinical profile against ER plus breast cancer

Summary: Breast cancer is a common malignant tumor in women, and drug resistance remains a clinical challenge. In this study, a novel compound, G-5b, was developed with potent antagonistic and degradation activities comparable to the current drug fulvestrant. G-5b also showed improved stability and solubility. Mechanistically, G-5b engages the proteasome pathway to degrade ER, inhibiting the ER signaling pathway and inducing apoptosis and cell cycle arrest. In animal models, G-5b exhibited superior pharmacokinetics and pharmacodynamics properties. Overall, G-5b is a promising long-acting SERD worthy of further investigation and optimization.

HDAC specificity and kinase off-targeting by purine-benzohydroxamate anti-hematological tumor agents

Summary: A series of hybrid inhibitors combining pharmacophores of known kinase inhibitors and benzohydroxamate HDAC inhibitors were synthesized and evaluated for their anticancer activity and pharmacokinetic properties. Compounds 4d-f exhibited promising cytotoxicity against hematological cells and moderate activity against solid tumor models. Compound 4d showed potent inhibition of multiple kinase targets and had stable interactions with HDAC and members of the JAK family. These compounds showed selective cytotoxicity with minimal effects on non-tumorigenic cells and favorable pharmacokinetic profiles.

Unexpected rearrangement of ivermectin in the synthesis of new derivatives with trypanocidal and antiplasmodial activities

Design and synthesis of pterostilbene derivatives bearing triazole moiety that might treat DSS-induced colitis in mice through modulation of NF-KB/ MAPK signaling pathways

Summary: In this study, a series of novel anti-inflammatory compounds were designed and synthesized based on the natural product pterostilbene skeleton. Among them, compound 8 showed the highest activity and exhibited its effects through inhibition of pro-inflammatory cytokines by blocking the NF-KB/MAPK signaling pathway. Compound 8 also demonstrated a good relieving effect on acute colitis in mice and showed good safety in acute toxicity experiments.
