Simultaneous quantitative determination, identification and qualitative screening of pesticides in fruits and vegetables using LC-Q-Orbitrap™-MS

A method based on QuEChERS extraction and LC-quadrupole-Orbitrap MS detection was established utilising an improved fully non-targeted way of data acquisition with and without fragmentation. A full-scan acquisition event without fragmentation (resolving power 70000) was followed by five consecutive fragmentation events (variable data independent acquisition - vDIA; resolving power 35000) where all ions from the full-scan range are fragmented. Compared with fragmentation in a single event (all-ion fragmentation - AIF), this improves both selectivity and sensitivity for the fragment ions, which is beneficial for screening performance and identification capability. The method was validated, using the data from the same measurements, for two types of analysis: quantitation/identification and qualitative screening. The quantitative validation, performed according to the guidelines in SANCO/12571/2013, tested the performance of the method for 184 compounds in lettuce and orange at two spiking levels: 10 and 50ngg(-1). The validation showed that the vast majority of the compounds met the criteria for trueness and precision set in the SANCO guidance document. In the qualitative validation the same 184 compounds were used to test the untargeted screening capabilities of the method. In this validation the compounds were spiked at three levels into 11 different fruit and vegetable matrices, which were measured twice on separate days. Taking all data from the qualitative validation together, an overall detection rate of 92% was achieved at the 10ngg(-1) level, increasing to 98% at 200ngg(-1). A screening detection limit (as defined in the SANCO guidelines) of 10ngg(-1) could be achieved for 134 compounds. For 39 and two pesticides the SDL was 50 and 200ngg(-1), respectively. For the other nine compounds no SDL could be established. The identification (ion ratio) criteria as recommended in the SANCO document could be met for 93% of the detected pesticide/matrix/concentration combinations. The outcome of both validations shows that the described method can be used to combine quantitative analysis and the identification of frequently detected pesticides (so far typically done using triple quadrupole MS/MS) with a qualitative screening to be used for a wide range of less frequent detected compounds in one measurement.

Influence of different hydrophilic interaction liquid chromatography stationary phases onmethod performance for the determination of highly polar anionic pesticides in complex feed matrices

Summary: Hydrophilic interaction liquid chromatography is recognized as an important solution for separating polar compounds not amenable to reverse phase chromatography. This study evaluated three different hydrophilic liquid chromatography stationary phases for the determination of 14 highly polar anionic molecules, showcasing different matrix effects and interactions between compounds. The use of isotopically labeled internal standards enabled acceptable quantitative performance and identification down to 0.02mgkg(-1) for most compounds.

Summary: The Human Biomonitoring for Europe initiative (HBM4EU) aims to study the exposure of citizens to chemicals and potentially associated health effects. In this project, interlaboratory comparison investigations and external quality assurance schemes were organized to ensure data consistency and reliability. The results showed that Europe has a strong analytical capability for bisphenol A (BPA) and bisphenol S (BPS), while improvements may be needed for bisphenol F (BPF) analysis.

Summary: This article presents the current advances and research needs in studying mycotoxins under the European Human Biomonitoring Initiative. The focus is on the mycotoxins deoxynivalenol (DON) and fumonisin B-1 (FB1), and their potential health impacts. Policy questions regarding hazard characterization, exposure, and risk assessment are addressed. The knowledge gained from this research is expected to support policy making and drive new Human Biomonitoring studies on mycotoxin exposure in Europe.

Summary: Polycyclic aromatic hydrocarbons (PAHs) were included in the European Human Biomonitoring Initiative (HBM4EU) for harmonization and advancement of human biomonitoring in Europe. A specific QA/QC program using ICIs and EQUASs was developed to ensure comparability and accuracy of analytical laboratories. The study found that 86% of participants achieved satisfactory results in determining 13 PAH metabolites in urine, although low limits of quantification were required. The HBM4EU QA/QC program established an international network of laboratories for comparable analysis of urinary PAH biomarkers.

Development and validation of two analytical strategies for the determination of glucosides of acidic herbicides in cereals and oilseed matrices

Summary: The aim of this research was to develop and validate a method for determining acidic herbicide glucosides, using enzymatic deconjugation as an alternative to alkaline hydrolysis. One specific enzyme was found to be practical and repeatable for deconjugation, leading to complete deconjugation of the glucosides. The method was validated according to established guidelines and a quantitative method for direct determination of intact glucosides was also optimized and validated.
