Involvement of CYP1B1 in interferon γ-induced alterations of epithelial barrier integrity

BACKGROUND AND PURPOSE CYP1B1 and CYP1A1 are important extra-hepatic cytochromes, expressed in the colon and involved in the metabolism of dietary constituents and exogenous compounds. CYP1B1 expression is increased by pro-inflammatory cytokines, and it has been recently implicated in regulation of blood brain barrier function. We investigated its involvement in the increased permeability of the intestinal epithelial barrier observed in inflammatory conditions. EXPERIMENTAL APPROACH Epithelial monolayers formed by human T84 colon carcinoma cells cultured on transwells, were disrupted by incubation with IFN gamma (10 ng.mL(-1)). Monolayer integrity was measured using transepithelial electrical resistance. CYP1A1 and CYP1B1 inhibitors or inducers were applied apically. Potential mechanisms of action were investigated using RT-qPCR. KEY RESULTS IFN gamma disrupts the barrier integrity of the T84 monolayers and increases CYP1B1 and HIF1 alpha mRNA expression. CYP1B1 induction is inhibited by the NF-kappa B inhibitor ammonium pyrrolidinedithiocarbamate (100 mu M) but not by the HIF1 alpha inhibitor 3-(5-hydroxymethyl-2-furyl)-1-benzyl indazole (50 mu M). Inhibition of CYP1B1 with the selective inhibitor 2,4,3,5-tetramethoxystilbene (100 nM) partly reverses the effects of IFN gamma on epithelial permeability. CONCLUSIONS AND IMPLICATIONS These data suggest that increased expression of CYP1B1 is involved in the effects of IFN gamma on epithelial permeability. Inhibition of CYP1B1 counteracts the alterations of epithelial barrier integrity induced by IFN gamma and could thus have a therapeutic potential in disorders of intestinal permeability associated with inflammation.

25-Hydroxycholesterol metabolism is altered by lung inflammation, and its local administration modulates lung inflammation in mice

Summary: Inflammation is a critical component of many lung diseases, and studies have shown that only one type of oxysterol levels change during lung inflammation, with different effects on its levels in different disease models. Experiments demonstrated that this oxysterol levels increased in the acute lung inflammation model, leading to a decrease in inflammatory cytokine secretion.

Effects of R-flurbiprofen and the oxygenated metabolites of endocannabinoids in inflammatory pain mice models

Summary: Pain is a cardinal sign of inflammation, with prostaglandins being major bioactive lipids involved in this process. The enzyme COX-2 not only synthesizes prostaglandins, but also metabolizes endocannabinoids to produce derivatives that may play a role in nociception. Further research on these endocannabinoid metabolites may provide insights into inflammatory pain mechanisms.

CEACAM5, KLK6, SLC35D3, POSTN, and MUC2 mRNA Analysis Improves Detection and Allows Characterization of Tumor Cells in Lymph Nodes of Patients Who Have Colon Cancer

Summary: This study compared the effectiveness of analyzing a selected group of biomarker mRNAs with routine methods for detecting lymph node metastases in colon cancer patients. The results showed that analyzing CEACAM5 and KLK6 mRNA levels improved the detection of tumor cells in lymph nodes, and increasing the tissue volume analyzed for CEACAM5 led to an increase in the number of positive nodes detected.

Prognostic Significance of GPR55 mRNA Expression in Colon Cancer

Summary: This study investigated the prognostic value of GPR55 in colon cancer. Combining GPR55 with CEA, CXCL16, or CXCL17 improved the discrimination between high and low expression groups. The GPR55/CEA ratio showed the best result, with a significant increase in mean survival time.

5'AMP-activated protein kinase: an emerging target of phytochemicals to treat chronic inflammatory diseases

Summary: Inflammation is a defensive response of the organism to various injuries. Chronic inflammatory diseases are challenging to treat, and phytochemicals may provide an interesting alternative approach. AMP-activated protein kinase plays an important role in regulating inflammatory responses. This review summarizes studies on phytochemicals that activate AMPK to improve chronic inflammatory disease models, and highlights factors to consider when investigating AMPK in the context of phytochemicals.

Lipid nanocapsules for the nose-to-brain delivery of the anti-inflammatory bioactive lipid PGD2-G

Summary: Prostaglandin D2-glycerol ester (PGD2-G) was used as a targeting agent for neuroinflammation, and lipid nanocapsules (LNC) were used as a safe vehicle to transport PGD2-G to the central nervous system (CNS). PGD2-G-loaded LNC (PGD2-G-LNC) effectively reduced pro-inflammatory cytokine expression in activated microglial cells and the olfactory bulb. Coating LNC's surface with the cell-penetrating peptide, transactivator of transcription (TAT), increased its accumulation in the brain and reduced the expression of pro-inflammatory chemokines in the CNS.

Summary: One fourth of colorectal cancer patients undergoing curative surgery will relapse, and lymph node metastasis is the most important prognostic factor. However, the current histopathological examination method is subjective and unable to provide information on tumor aggressiveness. The researchers developed a qRT-PCR test called ColoNode, which detects mRNA levels of specific biomarkers to estimate tumor cell load and aggressiveness. In a study of 196 colon cancer patients, ColoNode identified all patients detected by histopathology and an additional 9 patients. ColoNode was found to be a highly significant risk factor and surpassed histopathology in identifying patients at risk of relapse.
