Summary: Hyperhomocysteinemia (HHcy) is associated with thrombotic diseases and can increase platelet count. This study demonstrates that HHcy promotes megakaryocytes differentiation through the GH-PI3K-Akt axis and is independent of thrombopoietin (TPO). Melatonin administration effectively reverses Hcy-induced thrombopoiesis. This research provides new insight into the mechanisms by which HHcy confers an increased thrombotic risk.

Summary: This study aimed to investigate the effect of Ibrutinib and ferroptosis inducers on colorectal cancer treatment and its underlying mechanism. The upregulation of Nrf2 was found to be correlated with CRC progression and antioxidant proteins. Ibrutinib increased the sensitivity of CRC cells to ferroptosis inducers by inhibiting Nrf2.

Summary: This study investigated the effect of IGF-1 combined with OPN on neuronal axon growth and protein expression levels, and its underlying mechanism. The results showed that IGF-1 combined with OPN promoted axon growth through the IGF-1R/Akt/mTOR signaling pathway in lipid rafts, and the effect was superior to either agent alone. However, this effect was suppressed by the mTOR inhibitor rapamycin or the lipid raft cholesterol extraction agent M-beta-CD. Additionally, M-beta-CD significantly decreased the expression of p-IR. Western blot analysis revealed that the IGF-1 combined with OPN group had the highest expression levels of IR and p-IR in the neuronal lipid rafts. The enrichment of IR by IGF-1 combined with OPN was weakened, and p-IR was reduced when M-beta-CD was administered to the lipid rafts of neurons. This study demonstrated that IGF-1 combined with OPN promoted axon growth by activating the IGF-1R/Akt/mTOR signaling pathway in lipid rafts.

Summary: Transketolase (TKT), an important enzyme in the pentose phosphate pathway, plays a crucial role in maintaining ribose 5-phosphate levels and cell growth. Recent studies have shown that TKT is not only involved in liver cancer development but also in colorectal cancer progression. Abnormal upregulation of TKT expression is associated with poor prognosis in colorectal cancer, and TKT promotes colorectal cancer cell growth and metastasis by enhancing glycolysis through AKT phosphorylation.

Summary: The RNA-binding protein AUF1 plays a crucial role in modulating mTORC2/Akt signaling by promoting membrane localization of Akt and facilitating its phosphorylation. AUF1 also mediates the phosphorylation of the mTORC2-regulated metabolic enzyme glutamine fructose-6-phosphate amidotransferase 1. These findings have significant implications for the therapeutic targeting of cancers with upregulated mTORC2/Akt signaling.

Summary: BACE-1 plays a role in regulating the transition of microglia in Alzheimer's disease. Deficiency of BACE-1 leads to increased expression of certain transcription factors involved in the transition from homeostatic to highly phagocytic microglia. Similar transition-state microglia in AD brains are associated with decreased levels of BACE-1 expression.

Summary: MicroRNAs (miRNAs) have been extensively studied as circulating biomarkers for early diagnosis and prognosis of many human diseases. This study proposes a generic electrochemical strategy integrated with molecular beacon-like catalyzed hairpin assembly (mCHA) circuit and MOF@Au@G-triplex/hemin nanozyme for ultrasensitive detection of miR-721. With the assistance of the mCHA circuit, double-amplified nanozymes and sensing interface, this biosensor demonstrates a low detection limit and high specificity, providing a promising potential method for the early diagnosis of acute myocarditis.
