Summary: This study introduces a single-cell cytokine secretion analysis platform that integrates three-dimensional cell culture and primary oral squamous cell carcinoma tumor cell co-culture, which allows for the investigation of immune responses of human macrophages stimulated with lipopolysaccharide. The differential modulation effect of the tumor microenvironment on cytokine secretions was observed, with TNF-a secretion attenuated and IL-6 increased. The findings highlight the importance of the cell microenvironment in single-cell analysis and its impact on immune cell responses.
