Discovery of a small molecule targeting ULK1-modulated cell death of triple negative breast cancer in vitro and in vivo

UNC-51-like kinase 1 (ULK1) is well-known to initiate autophagy, and the downregulation of ULK1 has been found in most breast cancer tissues. Thus, the activation of ULK1-modulated autophagy could be a promising strategy for breast cancer therapy. In this study, we found that ULK1 was remarkably downregulated in breast cancer tissue samples by The Cancer Genome Atlas (TCGA) analysis and tissue microarray (TMA) analysis, especially in triple negative breast cancer (TNBC). To design a ULK1 agonist, we integrated in silico screening and chemical synthesis to acquire a series of small molecule candidates. After rounds of kinase and anti-proliferative activity screening, we discovered the small molecule, LYN-1604, to be the best candidate for a ULK1 agonist. Additionally, we identified that three amino acid residues (LYS50, LEU53, and TYR89) were key to the activation site of LYN-1604 and ULK1 by site-directed mutagenesis and biochemical assays. Subsequently, we demonstrated that LYN-1604 could induce cell death, associated with autophagy by the ULK complex (ULK1-mATG13-FIP200-ATG101) in MDA-MB-231 cells. To further explore LYN-1604-induced autophagic mechanisms, we found some potential ULK1 interactors, such as ATF3, RAD21, and caspase3, by performing comparative microarray analysis. Intriguingly, we found that LYN-1604 induced cell death involved in ATF3, RAD21, and caspase3, accompanied by autophagy and apoptosis. Moreover, we demonstrated that LYN-1604 has potential for good therapeutic effects on TNBC by targeting ULK1-modulated cell death in vivo; thus making this ULK1 agonist a novel potential small-molecule drug candidate for future TNBC therapy.

The planted GQDs show prolonged blood circulation and increased tumor accumulation compared to typical GQDs, and also enable long-term real-time visualization of the local pharmacokinetics of nanoparticles in vivo.

Highly active Pt/In2O3-ZrO2 catalyst for CO2 hydrogenation to methanol with enhanced CO tolerance: The effects of ZrO2

Summary: This study demonstrates high activity and enhanced CO tolerance for CO2 hydrogenation to methanol on Pt/In2O3-ZrO2 catalyst. The addition of ZrO2 enables stronger electron transfer between Pt and In2O3-ZrO2, leading to weaker CO adsorption and improved CO tolerance of Pt catalyst. The ZrO2-modified oxygen vacancy promotes CO2 activation.

Room-temperature high-speed mid-infrared quantum cascade laser with π-shape metal contact

By monolithically integrating a pi-shaped metal contact electrode, the parasitic capacitance of the device is reduced from 36.6 to 7.1 pF, resulting in an increased -3 dB RF modulation bandwidth from 870 MHz to 4.5 GHz compared to the conventional electrode configuration.

Methionine-Restricted Diet: A Feasible Strategy Against Chronic or Aging-Related Diseases

This review comprehensively summarizes the novel roles and interpretations of the mechanisms underlying MR in the prevention of chronic diseases and aging.

Heterogeneous grain size and enhanced hardness by precipitation of the BCC particles in medium entropy Fe-Ni-Cr alloys

Clonal Outbreak of Trichophyton tonsurans Causing Tinea Capitis Among a Wrestling Team in Beijing, China

Summary: An outbreak of tinea capitis caused by Trichophyton tonsurans was reported among five juvenile athletes in a wrestling team in Beijing, China. The fungal pathogens were identified as T. tonsurans by morphology and sequencing. Multilocus genotyping analysis revealed a single genotype among the five isolates. Antifungal susceptibility testing showed that these isolates were susceptible to common antifungal drugs for treating tinea capitis.

Shelf-life prediction and storage stability of Aeromonas bacteriophage vB_AsM_ZHF

Summary: Phage therapy is a potential alternative to antibiotics for bacterial infections. This study found that storing phages at 4 degrees C was optimal and that 10% dimethyl sulfoxide (DMSO) was more effective in protecting the phages at lower temperatures than glycerin. Additionally, the use of hydroxyapatite and a combination of trehalose with tryptic soy broth or skim milk powder alleviated phage inactivation during storage. Furthermore, a model for predicting phage shelf-life was established with high accuracy using the Accelerated Stability Assessment Program (ASAP) based on the Arrhenius equation.

How does ESG disclosure improve stock liquidity for enterprises - Empirical evidence from China

Identification of Balanol As a Potential Inhibitor of PAK1 That Induces Apoptosis and Cytoprotective Autophagy in Colorectal Cancer Cells

Summary: Colorectal cancer (CRC) is a common malignancy with poor prognosis and high incidence and mortality. PAK1 overexpression is associated with poor prognosis in colorectal cancer, and PAK1-targeted inhibition using a compound called Balanol is a potential therapeutic strategy. In vitro studies showed that Balanol effectively inhibits PAK1 and has anti-proliferative and anti-migration activity in SW480 cells. Balanol also induces apoptosis and cytoprotective autophagy in SW480 cells. These findings suggest that Balanol is a potential novel PAK1 inhibitor for future CRC treatment.

Cucurbituril-based supramolecular host-guest complexes: single-crystal structures and dual-state fluorescence enhancement

Engineering of cell-surface receptors for analysis of receptor internalization and detection of receptor-specific glycosylation

Summary: The epidermal growth factor receptor (EGFR) is a cell-surface glycoprotein involved in cell proliferation and tumor development. This study used a fluorescently labeled EGFR to investigate its time-dependent endocytosis in live cells and found that appended glycans affect EGFR internalization. Additionally, the study detected sialic acid residues attached to EGFR on the live cell surface using FRET-based imaging. This research provides valuable insights into the cellular functions of EGFR.

Ultrasensitive detection of aromatic water pollutants through protein immobilization driven organic electrochemical transistors

Summary: This study proposes a strategy that combines organic electronics with biosensor scaffolds to create a compact device for monitoring environmental aromatic pollution. Exclusive phenol detection with minimal loss of sensitivity could be achieved in complex pollutant mixtures and real environmental samples.

Selective FRET nano probe based on carbon dots and naphthalimide-isatin for the ratiometric detection of peroxynitrite in drug-induced liver injury

Summary: Drug-induced liver injury (DILI) is a common cause of acute liver failure in the USA and Europe, but most cases can be recovered or prevented by discontinuing the offending drug. Recent research has found that peroxynitrite (ONOO-) can be used as a potential indicator for early diagnosis of DILI, and there is an urgent need to establish a method to detect and track peroxynitrite in DILI cases. In this study, a FRET-based nano fluorescent probe CD-N-I was developed, which showed high selectivity and sensitivity in detecting peroxynitrite. The probe successfully detected exogenous peroxynitrite in live cells and endogenous peroxynitrite in APAP-induced liver injury of HepG2 cells.

Direct conversion of carboxylic acids to free thiols via radical relay acridine photocatalysis enabled by N-O bond cleavage

Strain induced reactivity of cyclic iminoboranes: the (2+2) cycloaddition of a 1H-1,3,2-diazaborepine with ethene

Compared to linear amino-iminoboranes, this cyclic iminoborane exhibits weakened bond strength and lower Lewis acidity value. The study suggests that the reduced ring strain of cyclic iminoborane prevents nitrogen fixation but allows facile (2 + 2) cycloaddition reaction with C2H4.
