Emerging Cytotoxic Alkaloids in the Battle against Cancer: Overview of Molecular Mechanisms

Considered as the second deadliest disease globally, cancer has captured the attention of researchers who have been trying with perseverance to decode its hidden aspects, to find new prognosis methods, and to develop better and more effective treatments. Plants have continuously offered an excess of unique secondary metabolites with remarkable biological applications. Alkaloids, one of the most abundant metabolites, constitute a large conglomerate of basic heterocyclic nitrogen-containing natural compounds which are normally produced by plants as toxic substances. Out of the 27,000 different alkaloids, more than 17,000 have displayed diversified pharmacological properties including anticancer activities. These metabolites have been classified either according to their chemical structures or their taxonomic origin. None of the researched alkaloids have been classified according to their molecular mechanism of action against cancer. In fact, only a fraction of the tremendous number of anticancer alkaloids has been copiously mentioned in journals. Here, we aim to provide a summary of the literature on some of the promising anticancer alkaloids that have not been well discussed previously and to classify them according to their molecular mechanisms of action. This review will provide a better understanding of the anticancer mechanisms of these promising natural products that are a rich reservoir for drug discovery.

Summary: The long-term success of standard anticancer monotherapeutic strategies has been limited by tolerability issues, treatment resistance, and cancer relapse. Combination therapy has shown promising results in strengthening the anti-tumor potential, reducing toxic effects, and overcoming resistance. Interest in combining natural products with conventional chemotherapeutics to improve treatment efficacy has been growing, with Thymoquinone (TQ) emerging as a promising candidate due to its low toxicity and anticancer efficacy.

Fucoidan and Alginate from the Brown Algae Colpomenia sinuosa and Their Combination with Vitamin C Trigger Apoptosis in Colon Cancer

Summary: This study investigated the antioxidant, cytotoxic, and apoptotic effects of fucoidan and alginate from brown algae Colpomenia sinuosa on cancer cell lines. The results showed that highly sulfated fucoidans had stronger antioxidant and cytotoxic effects than alginate. When combined with vitamin C, fucoidan exhibited enhanced inhibitory effects on colon cancer cells viability.

Thymoquinone Radiosensitizes Human Colorectal Cancer Cells in 2D and 3D Culture Models

Summary: This study demonstrates that combining thymoquinone (TQ) with ionizing radiation (IR) can enhance the sensitivity of colorectal cancer cells to radiotherapy, while sparing normal intestinal cells. TQ sensitization is associated with G2/M arrest, DNA damage, and changes in key signaling molecules. In addition, the combination treatment inhibits sphere formation and organoid forming ability. These findings suggest that combining TQ and IR could be a promising therapeutic strategy for eradicating colorectal cancer cells.

Micelles as potential drug delivery systems for colorectal cancer treatment

Summary: Colorectal cancer is still one of the most fatal malignancies worldwide despite significant progress in cancer therapy. The long-term effectiveness of chemotherapy drugs has been hindered by various limitations. In recent years, nanotechnology-based drug delivery systems, particularly polymeric micelles, have emerged as a promising approach to overcome these limitations. Polymeric micelles have shown great potential in improving the pharmacokinetic properties and safety profile of chemotherapeutic drugs, as well as achieving synergistic therapeutic outcomes through combination therapy.
