Activation of adenosine A2A receptor signaling regulates the expression of cytokines associated with immunologic dysfunction in BTBR T+ Itpr3tf/J mice

Autism spectrum disorder (ASD) is neurodevelopmental disorders characterized by stereotypical repetitive behavior, impaired social interaction, and deficits in communication. The BTBR T+ Itpr3(tf)/J (BTBR) mice have been extensively used as an animal model of the ASD-like phenotype. Adenosine A2A receptors (A2ARs) are considered potential targets in the treatment of neurodegenerative diseases. In this study, we used the A2AR antagonist SCH 5826 (SCH) and the A2AR agonist CGS 21680 (CGS) to investigate the activation of A2AR signaling in immune cells. Further, we examined the effects of A2ARs on the expression of the cytokines interleukin 2 (IL-2), IL-6, IL-9, interferon gamma (IFN-gamma), tumor necrosis factor alpha (TNF-alpha), and transforming growth factor beta (TGF-beta) in the spleen and in splenic CD4(+) T cells. In addition, we assessed the mRNA and protein expression levels of these cytokines in the brain tissue. Our results showed that the levels of IL-2(+), IL-6(+), IL-9(+), IFN-gamma(+), and TNF-alpha(+) were significantly lower, whereas the levels of TGF-beta(+) in the spleen and in splenic CD4(+) T cells were significantly higher in the CGS-treated mice than in the BTBR control and SCH-treated mice. In addition, reverse transcription polymerase chain reaction (RT-PCR) and western blot analysis showed a decrease in the mRNA and protein expression levels of IL-2, IL-6, IL-9, IFN-gamma(+), and TNF-alpha(+) and an increase in the mRNA and protein expression levels of TGF-beta in the CGS-treated mice, while treatment with BTBR alone and SCH resulted in increased Th1 levels and decreased Th2 levels in the brain tissue. Our results suggest that treatment the A2AR agonist CGS may be a promising therapeutic option for neuroimmune dysfunction. (C) 2017 Elsevier Inc. All rights reserved.

Mesoporous Silica Nanoparticles Coated with Carboxymethyl Chitosan for 5-Fluorouracil Ocular Delivery: Characterization, In Vitro and In Vivo Studies

Summary: This study developed amino-functionalized silica nanoparticles (AMSN) and O-Carboxymethyl chitosan-coated AMSN (AMSN-CMC) for ocular delivery of 5-Fluorouracil (5-FU). The optimal formulation of AMSN-CMC-FU showed improved corneal permeation and higher bioavailability compared to 5-FU gel. AMSN-CMC-FU gel was minimally irritating to rabbit eyes, suggesting its potential as a topical formulation for ocular cancer treatment.

CXCR3 antagonist NBI-74330 mitigates joint inflammation in Collagen-Induced arthritis model in DBA/1J mice

Summary: In this study, the CXCR3-specific antagonist NBI-74330 was used to inhibit T-cell-mediated signaling in a mouse model of rheumatoid arthritis (RA). The results showed that NBI-74330 treatment significantly reduced arthritis symptoms and histopathological changes. Additionally, NBI-74330 treatment downregulated the percentages and gene expression levels of Th1, Th17, and Th22 cells. These findings suggest that NBI-74330 has potential as a treatment for RA.

In this study, the effectiveness of an ITK inhibitor in reducing sepsis-induced neuroinflammation and depression-like symptoms in mice was investigated.

Summary: This study found that cadmium exposure exacerbates self-grooming and marble-burying behaviors, decreases social interactions, and leads to immune abnormalities in an autism spectrum disorder animal model. Further investigation revealed potential molecular mechanisms underlying these effects. These findings have important implications for the etiology of autism spectrum disorders and provide further evidence of heavy metals contributing to neurodevelopmental disorders through proinflammatory effects.

Summary: Autism spectrum disorder (ASD) is a childhood neuropsychiatric disorder that affects social skill and language development. This study explores the dysregulation of the redox couple TrxR1/Trx1 in T cells of ASD subjects, which is associated with decreased Bcl2 expression and increased apoptosis. The findings suggest that T cells in ASD subjects may have decreased survival during chronic inflammation. Future studies should investigate environmental factors that may contribute to abnormal immune responses in ASD subjects.

Summary: Autism spectrum disorder (ASD) is a complex disorder involving environmental, immune, and genetic factors. This study investigated the role of DNA methylation and the expression of DNMT1 in neutrophils of children with ASD. The study found that ASD subjects had reduced DNMT1 expression, DNA hypomethylation, and increased inflammatory mediators. The environmental pollutant DEHP further downregulated DNMT1 expression, highlighting its impact on epigenetic machinery in neutrophils of ASD subjects.

The results showed that the nanoparticles had stable physicochemical properties and demonstrated improved drug release, permeability, and pharmacodynamics compared to the aqueous solution of dexamethasone.
