The Importance of Ubiquitination and Deubiquitination in Cellular Reprogramming

Ubiquitination of core stem cell transcription factors can directly affect stem cell maintenance and differentiation. Ubiquitination and deubiquitination must occur in a timely and well-coordinated manner to regulate the protein turnover of several stemness related proteins, resulting in optimal embryonic stem cell maintenance and differentiation. There are two switches: an E3 ubiquitin ligase enzyme that tags ubiquitin molecules to the target proteins for proteolysis and a second enzyme, the deubiquitinating enzyme (DUBs), that performs the opposite action, thereby preventing proteolysis. In order to maintain stemness and to allow for efficient differentiation, both ubiquitination and deubiquitination molecular switches must operate properly in a balanced manner. In this review, we have summarized the importance of the ubiquitination of core stem cell transcription factors, such as Oct3/4, c-Myc, Sox2, Klf4, Nanog, and LIN28, during cellular reprogramming. Furthermore, we emphasize the role of DUBs in regulating core stem cell transcriptional factors and their function in stem cell maintenance and differentiation. We also discuss the possibility of using DUBs, along with core transcription factors, to efficiently generate induced pluripotent stem cells. Our review provides a relatively new understanding regarding the importance of ubiquitination/deubiquitination of stem cell transcription factors for efficient cellular reprogramming.

Clinical features and long-term treatment outcomes in choroidal tuberculoma

Summary: The clinical features and treatment outcomes of patients with choroidal tuberculoma were investigated. The study found that choroidal tuberculoma can develop and recur without evidence of systemic tuberculosis. Early detection and management, as well as positive results on immunological tests to initiate antitubercular therapy, are important.

Complications, treatments, and visual prognosis of choroidal osteomas

Summary: This retrospective study on choroidal osteoma aimed to report complications, treatments, and visual prognosis. Findings revealed that choroidal neovascularization (CNV) and subretinal fluid were the main visually significant complications, with intravitreal anti-VEGF injection being the common treatment choice. Results showed that the presence of CNV and outer retinal tubulation (ORT) were independent risk factors for vision loss, emphasizing the importance of regular check-ups and appropriate treatment options for better visual outcomes.

Pigmentary abnormality without significant drusen as a risk factor for late age-related macular degeneration

Summary: This study investigated the occurrence and risk factors of late age-related macular degeneration (AMD) in the fellow eye (FE) without significant drusen in patients with unilateral exudative macular neovascularization (MNV). The findings suggest that hypopigmentation on color fundus photography (CFP) and shallow irregular RPE elevation (SIRE) on OCT might represent the same lesion. Even small hypopigmentation and small RPE elevation were found to be significant risk factors for progression to exudative MNV.

Clinical features, treatment factors, and long-term outcomes of optic disc pit maculopathy

Summary: This study evaluated the clinical features, treatment factors, and long-term outcomes of optic disc pit maculopathy in Korean patients. Two peak incidence groups were identified, and the primary treatment choice did not significantly impact anatomical outcomes. However, delayed vitrectomy was associated with poorer visual outcomes.

Noncanonical Splice Site and Deep Intronic FRMD7 Variants Activate Cryptic Exons in X-linked Infantile Nystagmus

Summary: This study reports the discovery of noncoding pathogenic variants in patients with FRMD7-related infantile nystagmus (FIN). By using genome sequencing and targeted next-generation sequencing analysis, intronic variants in the FRMD7 gene were identified in patients with FIN. Through deep learning-based annotation and in vitro functional study, the impact of these variants on eye movements was confirmed, demonstrating the effectiveness of deep learning-based splicing tools in identifying hidden pathogenic variants in previously unsolved patients with infantile nystagmus.

Diagnostic Accuracy of Vitreous Cytology in Patients with Vitreoretinal Lymphoma

Clinical and Multimodal Imaging Features of Choroidal Nevi in the Korean Population

By developing a retinal pigment epithelium (RPE)-specific CRISPR/Cas9 system, we demonstrated its effectiveness in human retinal organoids and mouse models. This specific gene editing approach has potential applications in targeted therapy for specific diseases.

Summary: This study investigated the clinical features, natural course, and genetic characteristics of Koreans with rhodopsin-associated retinitis pigmentosa (RHO-associated RP). The study found that the generalized RP group had a higher rate of visual field impairment progression to late-stage RP compared to the sector RP group. Best-corrected visual acuity deterioration was observed only in the generalized RP group. The study also identified several RHO gene mutations, including some that presented with both generalized and sector RP phenotypes.

Therapeutic Efficacy of Spironolactone for Central Serous Chorioretinopathy
