Summary: Peripheral nerve injuries are a common cause of disabilities, and treatment with long nerve gap autografts is currently considered the best method. However, due to limited availability and potential issues at the donor site, research is focusing on developing regenerative strategies using artificial nerve guidance conduits (NGCs). These NGCs can be optimized through various methods to enhance nerve regeneration, and recent advancements in using different types of nanoparticles to introduce growth factors to NGCs are discussed in depth.

Summary: Growth-associated protein 43 plays a key role in neurite outgrowth through cytoskeleton remodeling. In this study, it was found that growth-associated protein 43 is expressed not only in the regenerating nerve tissue but also in the spinal cord after peripheral nerve transection. These findings suggest that growth-associated protein 43 could be used as a marker to monitor changes in the central nervous system after peripheral nerve injury.

Summary: The study assessed the efficacy of NH2-GO and Fr in a rat facial nerve motor neuron axotomy model using a ST conduit. Results showed an increase in regenerating axons and mean axon diameter with NH2-GO and a decrease in myelin surface area with Fr. Joint application of NH2-GO and Fr led to more regenerated axons and increased myelin thickness compared to the hydrogel group.

Summary: This study evaluated a biosimulated nerve repair system providing three types of regenerative cues in a peripheral nerve injury model. The nanofibers supported PC12 proliferation, preserved muscle mass, and gradually improved functional recovery. The combination of physical guidance, chemical cues, and therapeutic cues offers a promising strategy for the regeneration of severed peripheral nerves.

Summary: In this study, polydopamine-modified chitin conduits loaded with brain-derived neurotrophic factor mimetic peptides and vascular endothelial growth factor mimetic peptides were created. The conduits showed low cytotoxicity and sustained release of functional peptides. Application of the conduits in rat models of sciatic nerve injury improved motor function and promoted nerve regeneration.

Summary: Peripheral nerve injury is a common complication, and the traditional autologous nerve graft approach is limited. Nerve guidance conduits are widely used as an alternative, but they currently cannot meet the needs of large gap nerve injuries. Functional nerve conduits that incorporate electrical or magnetic stimulation and Schwann cells show promise for bridging large gap nerve injuries. Additionally, functional nerve conduits provide a new method for constructing drug and growth factor delivery systems in vivo.
