Summary: Nano-immunotherapy has shown potential in improving outcomes for triple negative breast cancer patients, but mechanisms of resistance need to be understood. By decompressing compressed blood vessels in lung metastases, perfusion can be restored and hypoxia alleviated, leading to more efficient delivery of nanomedicines and enhancement of anti-tumor immunity. Mechanotherapeutics in combination with nano-immunotherapy may offer synergistic effects on efficacy in breast cancer treatment.

Summary: Immunotherapy and anti-EGFR targeted treatment play a crucial role in colorectal cancer, but resistance remains a challenge. CXCL10 expression is associated with CD8+ T cell infiltration and tumor vascular normalization in colorectal cancer. CXCL10 overexpression inhibits tumor growth, increases CD8+ T cell infiltration, and induces tumor vascular normalization. CXCL10 interacts with VCAN to mediate tumor vascular normalization. Elevated CXCL10 expression enhances the sensitivity of colorectal cancer cells to cetuximab/anti-PD1 combination therapy. CXCL10 may be used to enhance the efficacy of anti-EGFR therapy and immunotherapy by targeting both tumor vessels and immune cells in colorectal cancer.

The nanomodulator effectively inhibits tumor neovascularization, normalizes blood vessels, and boosts immune response. This synergistic therapeutic strategy shows potential in suppressing the growth of TNBC in mice.

Summary: The PHD/HIF pathway is involved in immune and inflammatory processes, including in the hypoxic tumor microenvironment. Deleting PHD2 in T lymphocytes resulted in regression of EG7-OVA tumors, which depended on HIF-1a. The enhanced control of tumor growth was associated with increased polyfunctionality of CD8+ tumor-infiltrating lymphocytes, indicating the importance of glycolysis in sustaining CTL activity.
