Fructans from Agave tequilana with a Lower Degree of Polymerization Prevent Weight Gain, Hyperglycemia and Liver Steatosis in High-Fat Diet-Induced Obese Mice

Fructans from agave have received specific attention because of their highly branched fructan content. We have previously reported that the degree of polymerization (dp) influences their biological activity. Therefore, the aim of this study was to investigate the effect of unfractionated and fractionated fructans (higher and lower dps) from Agave tequilana in high-fat diet-induced (HFD) obese mice. Fructans with a lower dp (HFD+ScF) decreased weight gain by 30 %, body fat mass by 51 %, hyperglycemia by 25 % and liver steatosis by 40 %. Interestingly, unfractionated fructans (HFD+F) decreased glucose and triglycerides (TG), whereas fractionated fructans with a higher dp (HFD+LcF) decreased TG but not glucose; in contrast, HFD+ScF decreased glucose but not TG. Our findings suggest that both higher and lower dp agave fructans have complementary effects in metabolic disorders related to obesity. These findings may contribute to the development of improved food supplements with a specific ratio combination of fructans with different dps.

Summary: Multiple sclerosis is a chronic disease characterized by an autoimmune attack on axonal myelin sheaths in the central nervous system. Epigenetics has been studied as a research topic in the search for biomarkers and treatment targets for this disease. This study examined epigenetic marks in Peripheral Blood Mononuclear Cells (PBMCs) from MS patients treated with IFN-beta and GA or untreated, as well as healthy controls, and found that DNA methylation decreased in treated patients and correlated with clinical variables. However, no biomarker has been identified to predict treatment response before initiation.

Lipase B from Candida antarctica in Highly Saline AOT-Water-Isooctane Reverse Micelle Systems for Enhanced Esterification Reaction

Summary: The synthesis of butyl oleate by the lipase B from Candida antarctica (CalB) was studied in a reverse micellar system under extreme halophilic conditions. The impact of aqueous content and NaCl variation on the enzymatic activity of CalB in the butyl oleate reaction in reverse micelles was investigated. The results showed that increasing NaCl concentration led to higher enzymatic activity, reaching a maximum of 444.85 μmol/min at 5 M NaCl and Wo = 10, with pH 7.2 and 30 degrees C being the optimal conditions. However, the synthesis of butyl oleate by lipase CalB increased due to the reduction in the average reverse micelle size, as determined by dynamic light scattering (DLS). This study demonstrated the potential of reverse micelles as a system to enhance mass transport in heterogeneous biocatalysis and highlighted their promise for extreme halophilic lipases research.

Summary: The close interaction between fetal and maternal cells during pregnancy requires various immune-endocrine mechanisms to provide a tolerant environment for the fetus and protect it against infections. Prolactin (PRL), a hormone synthesized by the maternal decidua, is transported through the amnion-chorion and accumulates in the amniotic cavity, creating a hyperprolactinemic environment for the fetus. PRL has multiple immunomodulatory functions, but its precise role at the maternal-fetal interface is still not fully understood. This review summarizes the current knowledge on the immunological effects of PRL and its biological significance for the immune privilege of the maternal-fetal interface.

Summary: Autoimmune rheumatic diseases are a group of heterogeneous disorders characterized by clinical symptoms such as pain, tissue damage, immune deregulation, and inflammation. Biologic disease-modifying antirheumatic drugs are effective treatments, but their complexity can induce the development of anti-drug antibodies. TNF inhibitors, widely used in rheumatoid arthritis, appear to be the main contributors to immunogenicity. This review provides an overview of the immunogenicity of biological agents in autoimmune rheumatic diseases, focusing on the prevalence of anti-drug antibodies.
