Obstructive sleep apnea (OSA) associated chronic kidney disease ismainly caused by chronic intermittent hypoxia (CIH) triggered tissue damage. Receptor for advanced glycation end product (RAGE) and its ligand high mobility group box 1 (HMGB1) are expressed on renal cells and mediate inflammatory responses in OSA-related diseases. To determine their roles in CIH-induced renal injury, soluble RAGE (sRAGE), the RAGE neutralizing antibody, was intravenously administered in a CIH model. We also evaluated the effect of sRAGE on inflammation and apoptosis. Rats were divided into four groups: (1) normal air (NA), (2) CIH, (3) CIH+sRAGE, and (4) NA+sRAGE. Our results showed that CIH accelerated renal histological injury and upregulated RAGE-HMGB1 levels involving inflammatory (NF-kappa B, TNF-alpha, and IL-6), apoptotic (Bcl-2/Bax), and mitogen-activated protein kinases (phosphorylation of P38, ERK, and JNK) signal transduction pathways, which were abolished by sRAGE but p-ERK. Furthermore, sRAGE ameliorated renal dysfunction by attenuating tubular endothelial apoptosis determined by immunofluorescence staining of CD31 and TUNEL. These findings suggested that RAGE-HMGB1 activated chronic inflammatory transduction cascades that contributed to the pathogenesis of the CIH-induced renal injury. Inhibition of RAGE ligand interaction by sRAGE provided a therapeutic potential for CIH-induced renal injury, inflammation, and apoptosis through P38 and JNK pathways.

Summary: The study found that MSC SLP significantly promoted cell survival, inhibited epithelial cell apoptosis, attenuated inflammatory cell recruitment, and reversed immunological imbalances induced by bleomycin in ALI. MSC SLP ameliorated ALI by activating p63 and promoting p63(+) cell proliferation and the repair of damaged epithelial cells. The findings suggest that MSC SLP shows considerable therapeutic promise for treating ALI and ARDS.

Summary: This study examined the clinical phenotypic characteristics of older patients with newly diagnosed obstructive sleep apnea (OSA) and found that older patients displayed more symptoms and comorbidities associated with OSA, such as nocturia and decreased sleep quality. Additionally, being aged 65 years or older increased the risk of developing hypertension, coronary artery disease (CAD), and ischemic stroke.

Transcriptional Characterization of Bronchoalveolar Lavage Fluid Reveals Immune Microenvironment Alterations in Chemically Induced Acute Lung Injury

Summary: This study performed single-cell RNA sequencing on bronchoalveolar lavage fluid samples from phosgene-induced acute lung injury rat models and healthy controls. The results showed that dendritic cells and specific macrophage subclusters exhibited increased function during the early stage of pulmonary tissue damage. Nine subpopulations were identified that play multiple functional roles, including immune responses, pulmonary tissue repair, cellular metabolic cycle, and cholesterol metabolism. Pseudo-time trajectory analysis also suggested that proliferating macrophage clusters exerted multiple functional roles.
