Somatic Mutations in Cardiovascular Disease

Summary: Advances in genomic sequencing have increased our understanding of the inherited basis of cardiovascular disease (CVD). Reanalysis of genomic datasets revealed somatically acquired DNA mutations as an unexpected risk factor for CVD. This review provides an overview of somatic mutations and their contributions to CVD, focusing on the most common manifestation of clonal hematopoiesis. The available data on how somatic mutations lead to tissue mosaicism in various forms of CVD are also reviewed, along with future research directions and technological advances.

Summary: This study investigated the relationship between CHIP, LTL, and CAD using data from the Trans-Omics for Precision Medicine (TOPMed) program and UK Biobank. The results showed that genetically imputed longer LTL increased the risk of developing CHIP, which then hastened the shortening of measured LTL. There was also evidence of mediation between CHIP and CAD through LTL.

Lymphoid clonal hematopoiesis: implications for malignancy, immunity, and treatment

Summary: Clonal hematopoiesis (CH) is the expansion of hematopoietic stem cell clones due to acquired somatic mutations or mosaic chromosomal alterations. While somatic mutations are associated with myeloid malignancies, a recent study discovered a subset of mutations and chromosomal alterations that are specifically associated with lymphoid malignancies, termed lymphoid clonal hematopoiesis (L-CH). Unlike myeloid clonal hematopoiesis, L-CH can originate from both stem cells and differentiated progeny stages. This review explores the definition of L-CH in the context of lymphocyte maturation and lymphoid malignancy precursor disorders, its association with late-onset autoimmunity and immunodeficiency, as well as therapy-related L-CH following chemotherapy or hematopoietic stem cell transplantation.

Summary: This study aimed to investigate the presence of clonal hematopoiesis of indeterminate potential (CHIP) in young women treated for breast cancer and its impact on prognosis. The results showed that only a small number of patients had clones large enough to be diagnosed as CHIP, and there was no significant association between their prognosis and non-breast cancer-related outcomes.
