Incorporating an emulsion onto a gel base creates emulgel, a drug delivery system that enhances stability and penetration, allowing it to envelop both hydrophilic and lipophilic molecules. Emulgel provides controlled drug release with improved patient compliance and therapeutic efficacy. The preparation involves three main stages: creating a water-in-oil or oil-in-water emulsion, providing the gel base, and incorporating the emulsion onto the gel base with continuous stirring. Various materials are used, including different oils, emulsifiers, and gelling agents. The physical properties, particle size distribution, spreadability, permeation, and drug release rate are evaluated during development and characterization. Emulgel has potential clinical uses for treating skin disorders and diseases such as chronic anal fissures, and has been studied for delivering anti-acne, analgesic, and anti-inflammatory drugs.
Incorporation of an emulsion onto a gel base develops a drug delivery system with improved characteristics, known as emulgel, that can envelop both hydrophilic and lipophilic molecules, and therefore increase stability and penetration of topical formulations. Such a drug delivery system provides controlled drug release that has more patient compliance and higher therapeutic efficacy. Emulgel is prepared in three main stages, preparation of water-in-oil or oil-in-water emulsion, providing the gel base, and incorporation of prepared emulsion onto gel base with continuous stirring. Various materials such as different oils (e.g. sesame oil, balsam oil, and mineral oil), emulsifiers (e.g. Tween (R) and Span (R) as the non-ionic surfactant, polyvinyl alcohol), and gelling agents including cellulose derivatives such as hydroxypropyl methylcellulose (HPMC), hydroxyethylcellulose (HEC) and carboxymethyl cellulose (CMC) in different concentrations are used in emulgel preparation. The physical properties, particle size distribution, spreadability, permeation, and drug release rate are evaluated in their development and characterization. They are used in skin disorders and other diseases such as chronic anal fisher. Also, anti-acne, analgesic, and anti-inflammatory drugs have been formulated as emulgel delivery system and their effects have been studied. In this article, the subject is to review the characteristics, preparation methods, and therapeutic efficacy as well as the potential clinical use of emulgels.

Analysing the Effect of Permeation Enhancers on the In Vitro Skin Permeability of Propranolol: A Mechanistic Study

Eskandar Moghimipour, Arash Tajmal, Hossein H. E. I. D. A. R. I. Kaydan, Anayatollah Salimi

Summary: In this study, the effect of different permeation enhancers on the skin permeability of propranolol was analyzed. It was shown that all the enhancers improved the permeability of the drug through rat skin. Labrafac had the greatest effect on flux, with an ERflux value of 9.24, while Plurol oleique had the greatest effect on ERD, with a value of 13.99. Chemical permeation enhancers can be considered as suitable excipients in transdermal delivery of propranolol due to their ability to change the skin structure and increase drug permeability.

Absorption enhancer approach for protein delivery by various routes of administration: a rapid review

Toktam Salehi, Mohammad Amin Raeisi Estabragh, Soodeh Salarpour, Mandana Ohadi, Gholamreza Dehghannoudeh

Summary: Peptides and proteins play crucial roles in living organisms and their malfunction can lead to various diseases. The use of proteins in treating diseases like cancers and hepatitis is increasing. However, the large and hydrophilic structure of proteins and the presence of biological and lipophilic membranes pose limitations to protein administration. Increasing protein absorption through these membranes can be achieved by using absorption enhancers, such as surfactants, phospholipids, and cyclodextrins. This review focuses on other absorption enhancers and their mechanisms in protein administration routes.

Effects of aging and diabetes on the deformation mechanisms and molecular structural characteristics of collagen fibrils under daily activity

Wei-Han Hui, Yen-Lin Chen, Shu-Wei Chang

Summary: Crosslinking affects the mechanical behavior and tissue metabolism of collagen-based tissues. Aging and diabetes alter the crosslinking type and density, thus changing tissue properties. High-connectivity enzymatic crosslinking and advanced-glycation end-products result in uniform deformation under daily activity, while low-connectivity enzymatic crosslinking does not. High-connectivity model displays more sliding, while AGEs induce instability in structures near the binding sites.
