Summary: Researchers have identified a new subtype of lymphatic endothelial cells called iLECs, which are positive for Ptx3 and are involved in PI3K-driven lymphatic malformations. These cells recruit pro-lymphangiogenic macrophages and promote pathological lymphatic vessel growth. Understanding the specific disease pathogenesis of different vessel types could lead to the development of more efficient therapies.

Summary: Loss of PRC2 has a critical role in driving metastasis in malignant peripheral nerve sheath tumors (MPNSTs), leading to increased collagen-dependent invasion, upregulation of matrix-remodeling enzymes, and elevated lung metastasis. Clinical sample analysis confirms that PRC2 loss is associated with metastatic disease, increased fibrosis, and decreased survival in MPNST patients.

Summary: This study found a close correlation between TYMS and invasion ability in colon cancer cells, as well as its crucial role in EMT regulation. It suggests that chemotherapeutics targeting TYMS expression may enhance the effectiveness of colon cancer treatment, especially in the metastatic stage.

Summary: The study identified a module containing 151 genes that was significantly correlated with lymphatic invasion in colon cancer, with DAPK3 identified as the pseudohub of the module. High expression of DAPK3 was significantly associated with greater lymphatic invasion and poor overall survival. Gene ontology analysis suggested that the module is involved in tumor cell survival, migration, and epithelial-mesenchymal transformation.

Summary: This retrospective study investigated the characteristics of gastroenteropancreatic neuroendocrine tumors using a unified classification. Tumor size, depth, lymphatic invasion, venous invasion, and neuroendocrine tumor grade were significantly correlated with metastasis. Venous invasion was found to be more strongly correlated with metastasis than lymphatic invasion. These findings highlight the importance of a unified classification and standardized assessment framework in neuroendocrine tumor research.

Summary: This study aimed to determine whether the location of lymphatic invasion, vascular invasion, and perineural invasion could affect the survival of patients with stage II colon cancer. The results showed that extramural invasion was associated with a worse prognosis, while intramural invasion was not. Therefore, pathologic reports about the location of invasion may be helpful for predicting prognosis and determining the need for adjuvant chemotherapy in stage II colon cancers.

Summary: SUV420H2 is an important regulator of epithelial cell plasticity and its loss contributes to right-sided colorectal tumorigenesis through epigenetic regulation of chromatin compaction. Reduced levels of H4K20me3 and increased chromatin accessibility are observed in patient-derived right-sided colorectal cancer organoids. Inhibition of histone demethylases can lead to re-compaction of chromatin and selective growth inhibition in right-sided colorectal tumors.

Summary: The E3 ligase TNF receptor-associated factor 4 (TRAF4) is increased in colorectal cancer (CRC), and its depletion impairs CRC cell phenotype and enhances radiation-induced cell death. Irradiation activates the JNKs/c-Jun signaling, contributing to radioresistance by activating the antiapoptotic protein Bcl-xL. TRAF4 is positively correlated with c-Jun and Bcl-xL, and targeting the TRAF4/Bcl-xL axis sensitizes CRC cells to radiotherapy.

Summary: The study demonstrates that breast cancer brain metastasis cells undergo epithelial-to-mesenchymal transition (EMT) when infiltrating the brain parenchyma, and removing these infiltrated tumor cells can improve surgical efficacy, providing new avenues for patient treatment.

Summary: This study reveals a novel mechanism of collective metastasis in breast cancer, where tumor-associated macrophages (TAMs) induce the conversion of cancer cell clusters to an invasive state with high CD44 expression. The activation of the MDM2/p53 pathway by TAMs-released CCL8 mediates the acquisition of the CD44(high) state, leading to cohesive detachment and spread.

Summary: The expression of S100A4 in lymphatic endothelial cells promotes lymphatic vessel sprouting in growing tumors by regulating glycolysis. S100A4 plays a crucial role in the position and motility of tip cells, and its expression is upregulated under hypoxic conditions. These findings suggest that S100A4 is a novel regulator of tumor-induced lymphangiogenesis.

Summary: This study discovered that HIFs induce the expression of ADAM12, leading to EGFR signaling activation and subsequently promoting breast cancer cell migration, invasion, and metastasis. Inhibition of ADAM12 expression or activity can effectively reduce tumor cell migration, invasion, and lung metastasis.

Summary: IRF2BP2 is associated with lymph node metastasis in oral squamous cell carcinoma (OSCC). It promotes invasion and epithelial-mesenchymal transition (EMT) by regulating fatty acid oxidation (FAO) and mitochondrial fission. Targeting IRF2BP2-FAO may serve as a potential therapeutic strategy for LNM+ OSCC.
