In this study, researchers found that the small molecule HIF-1a inhibitor PX-478 inhibits the secretion of IL-1 beta, leading to decreased liver injury and improved ALF in mice. The administration of PX-478 represents a promising therapeutic strategy for inflammation-induced ALF.

Combined treatment of androgen deprivation therapy (ADT) with anti-IL-1 beta antibody and immune checkpoint inhibitor anti-PD-1 antibody shows a stronger anticancer effect on castration-resistant PCa.

Summary: Interleukin 1 beta (IL-1 beta) plays a crucial role in inflammation and is regulated by the inflammasome. Stimulation of LPS-primed human macrophages with sub-lytic levels of the membrane attack complex (MAC) can activate the NLRP3 inflammasome and trigger IL-1 beta release. These findings demonstrate the involvement of MAC in inflammasome activation and IL-1 beta release in human macrophages.

Summary: Autophagy in macrophages plays a role in reducing atherosclerosis by promoting lipid metabolism. Beta-arrestin-1, a member of the beta-arrestin family, is involved in this process. The study found that knocking out beta-arrestin-1 in myeloid cells promotes plaque formation, while overexpression of beta-arrestin-1 has the opposite effect. The protective effects of macrophage beta-arrestin-1 against atherosclerosis are mediated by autophagy.
