Small Molecule Inhibition of Rab7 Impairs B Cell Class Switching and Plasma Cell Survival To Dampen the Autoantibody Response in Murine Lupus

IgG autoantibodies mediate pathology in systemic lupus patients and lupus-prone mice. In this study, we showed that the class-switched IgG autoantibody response in MRL/Fas(lpr/lpr) and C57/Sle1Sle2Sle2 mice was blocked by the CID 1067700 compound, which specifically targeted Ras-related in brain 7 (Rab7), an endosome-localized small GTPase that was upregulated in activated human and mouse lupus B cells, leading to prevention of disease development and extension of lifespan. These were associated with decreased IgG-expressing B cells and plasma cells, but unchanged numbers and functions of myeloid cells and T cells. The Rab7 inhibitor suppressed T cell-dependent and T cell-independent Ab responses, but it did not affect T cell-mediated clearance of Chlamydia infection, consistent with a B cell-specific role of Rab7. Indeed, B cells and plasma cells were inherently sensitive to Rab7 gene knockout or Rab7 activity inhibition in class switching and survival, respectively, whereas proliferation/survival of B cells and generation of plasma cells were not affected. Impairment of NF-kappa B activation upon Rab7 inhibition, together with the rescue of B cell class switching and plasma cell survival by enforced NF-kappa B activation, indicated that Rab7 mediates these processes by promoting NF-kappa B activation, likely through signal transduction on intracellular membrane structures. Thus, a single Rab7-inhibiting small molecule can target two stages of B cell differentiation to dampen the pathogenic autoantibody response in lupus.

Characterization of Pathogenic CD8+ T cells in Chlamydia-Infected OT1 Mice

Summary: This study validated the critical role of CD8(+) T cells in the induction of hydrosalpinx caused by Chlamydia trachomatis. CD8(+) T cells from wild type C57BL/6J mice were able to promote hydrosalpinx in OT1 mice through transfer. However, this promotion needed to be initiated within 2 weeks after infection, and once initiated, CD8(+) T cells could continue to promote hydrosalpinx for more than 3 weeks.

Global, regional and national burden of gastroesophageal reflux disease, 1990-2019: update from the GBD 2019 study

Summary: This study evaluated the latest epidemiologic patterns and trends for GERD using data from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2019. The results showed that the burden of GERD is continuing to worsen, with a higher prevalence among females compared to males. Higher socio-demographic index (SDI) was associated with lower incidence and years of life lived with disability (YLDs) for GERD in 2019.

Interleukin-27 (IL-27) Promotes chlamydial Infection in the Female Genital Tract

Summary: In this study, it was found that IL-27 may promote chlamydial infection in the female mouse genital tract and increase chlamydial burden by suppressing IFN-gamma-mediated immunity. Knocking out IL-27R from T cells alone significantly shortened the infectious shedding courses of Chlamydia in the mouse genital tract. These findings suggest that attenuating IL-27 signaling in T cells may be used to enhance genital tract immunity against chlamydial infection.

Type-I Interferon Signaling Protects against Chlamydia trachomatis Infection in the Female Lower Genital Tract

Summary: We found that Chlamydia trachomatis is inhibited during early infection in the female mouse genital tract and that the innate immunity against C. trachomatis is compromised without cGAS-STING signaling. We evaluated the impact of type-I interferon signaling on C. trachomatis infection in the female genital tract. Our results showed that mice lacking the type-I interferon receptor (IFN alpha R1) had significantly higher levels of live chlamydial organisms on days 3 and 5, demonstrating the protective role of type-I interferon signaling in preventing C. trachomatis infection in the mouse genital tract.

Summary: According to the global burden of disease study, digestive diseases pose a significant burden worldwide and vary by age, sex, sociodemographic index, and geographical region. Cirrhosis and other chronic liver diseases are the leading contributors to the burden of digestive diseases globally. The age-standardized rate of digestive disease DALYs has decreased from 1990 to 2019, and alcohol use is the largest contributor to the global burden of digestive diseases.
