New Insights Into the Activity of Apple Dihydrochalcone Phloretin: Disturbance of Auxin Homeostasis as Physiological Basis of Phloretin Phytotoxic Action

Apple species are a unique source of dihydrochalcones, which have a mysterious role in plants and are related to apple replant disease (ARD). This study elucidated the physiological basis of the phytotoxic action of dihydrochalcone phloretin in Arabidopsis and identified it as a prospective eco-friendly phytotoxic compound.
Apple species are the unique naturally rich source of dihydrochalcones, phenolic compounds with an elusive role in planta, but suggested auto-allelochemical features related to apple replant disease (ARD). Our aim was to elucidate the physiological basis of the phytotoxic action of dihydrochalcone phloretin in the model plant Arabidopsis and to promote phloretin as a new prospective eco-friendly phytotoxic compound. Phloretin treatment induced a significant dose-dependent growth retardation and severe morphological abnormalities and agravitropic behavior in Arabidopsis seedlings. Histological examination revealed a reduced starch content in the columella cells and a serious disturbance in root architecture, which resulted in the reduction in length of meristematic and elongation zones. Significantly disturbed auxin metabolome profile in roots with a particularly increased content of IAA accumulated in the lateral parts of the root apex, accompanied by changes in the expression of auxin biosynthetic and transport genes, especially PIN1, PIN3, PIN7, and ABCB1, indicates the role of auxin in physiological basis of phloretin-induced growth retardation. The results reveal a disturbance of auxin homeostasis as the main mechanism of phytotoxic action of phloretin. This mechanism makes phloretin a prospective candidate for an eco-friendly bioherbicide and paves the way for further research of phloretin role in ARD.

Impairment of root auxin-cytokinins homeostasis induces collapse of incompatible melon grafts during fruit ripening

Summary: The collapse of certain melon-pumpkin graft combinations may be caused by impairment of photoassimilates transport and imbalance of root auxin and cytokinins. Incompatible combinations show elevated levels of IAA and certain cytokinins in the roots during fruit ripening, while restricting transport of photoassimilates and inducing their accumulation in the scion.

Phytohormones in Sweet Cherry Buds During Winter Rest and Bud Development

Summary: This study was conducted to assess the involvement of gibberellins (GAs), cytokinins (CKs), and auxins (AX) in the dormancy of sweet cherry buds. The results showed that these hormone groups did not exhibit significant changes during the transition between different dormancy phases. However, there were specific changes in the content of bioactive molecules during ontogenetic development.

RALF1 peptide triggers biphasic root growth inhibition upstream of auxin biosynthesis

Summary: This study investigates the mechanism of rapid alkalinization factors (RALFs) in regulating plant growth. It is found that RALF1 inhibits the growth of Arabidopsis thaliana roots by inducing apoplast alkalinization, which is mediated by the receptor FERONIA (FER) through net H+ influx. Additionally, the study reveals cross-talk between RALF1 and auxin signaling pathways, where RALF1 up-regulates auxin biosynthesis to regulate root growth. The biphasic action of RALF1 on growth allows plants to respond rapidly to environmental stimuli and reprogram growth and development in the long term.

Solanum dulcamara L. Berries: A Convenient Model System to Study Redox Processes in Relation to Fruit Ripening

Summary: This study provides a characterization of the redox processes associated with the ripening of bittersweet berries for the first time. Reactive oxygen species were measured using Electron Paramagnetic Resonance Spectroscopy and Imaging, and the metabolic profiling of antioxidants and antioxidant enzyme activity was assessed. Changes in antioxidants and oxidation processes occurred during the transition from green to ripe stage. The skin of bittersweet berries had the highest antioxidant potential, and the enzymatic antioxidant system played a critical protective role. The EPRI methodology was presented to assess the redox status of fruits and discriminate between different tissues. The intracellular reoxidation of nitroxide probe 3CP was observed for the first time and proposed as an indicator of oxidative stress during ripening. The noninvasive EPRI technique has the potential for broader application in studying redox processes in fruits and other oxidative stress-related circumstances.

Summary: In this study, an efficient tissue culture system for improving the rocket cultivar Domaca rukola was established. Optimal shoot multiplication and organogenesis protocols were determined through experiments with different hormone concentrations and combinations. In addition, in vitro-produced rocket shoots were found to be a potential source of the antioxidant flavonoid kaempferol.

Phytohormone Profiling of Potato (Solanum tuberosum L.) Exposed to French Marigold (Tagetes patula L.) Essential Oil

Summary: The study investigated the comprehensive phytohormone profile of potato plants exposed to French marigold essential oil (EO) for different time periods. The results showed altered phytohormone responses of EO-exposed potato plants, with stress-related phytohormones being the most affected. The expression levels of genes involved in biosynthesis and catabolism of stress-related phytohormones indicated that the jasmonic acid biosynthetic pathway was the most affected.
