In Vitro Gut Fermentation of Whey Protein Hydrolysate: An Evaluation of Its Potential Modulation on Infant Gut Microbiome

This study evaluated the potential effect of whey protein hydrolysate (WPH) on the infant gut microbiome. The results showed that WPH had a positive modulation on the gut microbiota, increasing the production of short-chain fatty acids (SCFA) and promoting the growth of probiotics. This could benefit the energy balance and metabolism of infants.
Whey protein and its hydrolysate are ubiquitously consumed as nutritional supplements. This study aimed to evaluate the potential effect of whey protein hydrolysate (WPH) on the infant gut microbiome, which is more variable than that of adults. Colonic fermentation was simulated through a static digestion model and fecal culture fermentation, using feces from normal infants aged from 1-3 years old. During in vitro gut fermentation, measurements of short-chain fatty acids (SCFA) concentrations and 16S rRNA amplicon sequencing were performed. Additionally, the growth curves of cultivated probiotics were analyzed to evaluate the prebiotic potential of WPH. Besides the decline of pH in fermentation, the addition of WPH induced a significant increase in the SCFA production and also the relative abundance of Proteobacteria, Bacteroides, and Streptococcus (p < 0.05). The lower ratio of Firmicutes/Bacteroidetes in WPH-supplemented samples indicated the positive modulation of WPH on the gut microbiota, which could benefit the energy balance and metabolism of infants. The stimulation effect of WPH on the probiotics (particularly Lactobacillus acidophilus NCFM) during cultivation implied the prebiotic potential as well. Our findings shed light on WPH as a valuable dietary supplement with not only enriched resources of essential amino acids but also the potential to restore the infant gut microbiome.

COS-RP produced by AsA/H2O2 redox pair showed higher antioxidant and antimicrobial activities compared to COS-HP, with lower degree of polymerisation and average molecular weight.

Silver carp (Hypophthalmichthys molitrix) by-product hydrolysates: A new nitrogen source for Bifidobacterium animalis ssp. lactis BB-12

Summary: This study examined the effect of silver carp by-product hydrolysates on the growth of Bifido-bacterium animalis ssp. lactis BB-12. Papain was found to recover more nitrogen and < 1 kDa components from the by-products compared to neutrase and alcalase. Papain-2 h, hydrolysates obtained by papain for 2 hours, showed greater effectiveness in promoting the growth of BB-12. The content of < 1 kDa components, sulfhydryl-containing amino acids, and peptides composed of all the hydrophobic amino acids in the hydrolysates were positively correlated to the growth of BB-12. BB-12 cultured with papain-2 h substitute exhibited a thinner and shorter morphology after 4 hours of cultivation. This study provides a new nitrogen source for Bifidobacterium animalis ssp. lactis BB-12 and suggests a possible strategy for utilizing by-products to reduce aquacultural waste.

Amino acid degradation and related quality changes caused by common spoilage bacteria in chill-stored grass carp (Ctenopharyngodon idella)

Summary: In this study, three major spoilage bacteria of grass carp were artificially inoculated in amino acid solutions and grass carp flesh to investigate their involvement in amino acid degradation and fish quality deterioration. The results showed that Pseudomonas putida highly degraded free amino acids and produced ammonia in grass carp flesh, while Aeromonas rivi-pollensis produced 3-Methyl-butanol and 2-Methyl-butanol through leucine and isoleucine degradation. Shewa-nella putrefaciens showed potent ornithine-decarboxylation activity and released putrescine. Additionally, when S. putrefaciens cooperated with P. putida, more putrescine was produced through the arginine deiminase pathway. In conclusion, this study provided fundamental information on the spoilage mechanisms of freshwater fish and important guidance for quality control strategies.

Oxidation affects dye binding of myofibrillar proteins via alteration in net charges mediated by a reduction in isoelectric point

Summary: This study investigated the effect of oxidation on the dye-binding behavior of myofibrillar proteins. Selected dyes with different charges were incubated with myofibrils oxidized by the Fenton system. The results showed that oxidation increased the particle size, hydrophobicity, and absolute value of Zeta-potential of myofibrillar proteins, while decreasing the net positive charges. The binding ability of myofibrillar proteins towards certain dyes was affected by the pH environment, suggesting that oxidation-induced change in net charges is the driving force affecting the amount of protein-bound dye.

Protein hydrolysate from salmon frame debittered by plastein reaction: amino acid composition, characteristics and antioxidant activities

Summary: This study investigated the effects of plastein reaction on the bitterness, physicochemical properties, and antioxidant activity of salmon frame hydrolysate using different proteases at varying concentrations and temperatures. The results showed that the plastein reaction significantly reduced the bitterness of the hydrolysate and improved its solubility, although there was a slight decrease in antioxidant activity.

Improvement of Spatial Memory and Cognitive Function in Mice via the Intervention of Milk Fat Globule Membrane

Summary: This study investigated the effects of milk fat globule membrane (MFGM) intervention on spatial memory and cognitive function in mice. The results showed that mice in the M group had more disordered trajectories in the Morris water maze experiment, and immunohistochemical analysis revealed a higher number of neurons in the hippocampal dentate gyrus (DG) area of the M group compared to the control group. Differentially expressed proteins (DEPs) related to synaptic formation and neurotransmitter receptors were also identified. The findings provide new insights into the potential mechanisms of spatial memory and cognitive enhancement by MFGM.

Whey Protein Hydrolysate Exerts Anti-Inflammatory Effects to Alleviate Dextran Sodium Sulfate (DSS)-Induced Colitis via Microbiome Restoration

Summary: The study found that whey protein hydrolysate (WPH) can prevent the development of colitis, protect the colonic mucosal layer, and reduce inflammation in mice. High doses of WPH had a positive impact on the microbiome, reducing harmful bacterial strains. This research suggests that WPH has anti-inflammatory properties and shows promise in managing colitis.
