Summary: This article discusses the current research on using siRNA-based gene silencing strategies to treat glaucoma, including discussions on ocular structures, administration routes, viral and non-viral vectors, as well as basic strategies and challenges in siRNA therapy. Different therapeutic targets for glaucoma, current siRNA-based drugs used in treatment, and strategies for siRNA-based treatment after glaucoma surgery are also briefly mentioned.

Summary: RNA interference (RNAi) mediated by siRNA-3WJ, which is a fusion of siRNA with the phi29 3WJ motif, can enter the RNAi pathway through Dicer cleavage and achieve gene silencing. It has been found that siRNA-3WJs are effective even in Dicer-deficient or Argonaute 2-deficient cells, and they work best when the antisense strand of the siRNA duplex is positioned 5' of the 3WJ.

Summary: A method to inhibit autophagy by altering the intracellular delivery fate of siRNA was designed. By adsorbing KDEL-grafted chondroitin sulfate (CK) onto cationic liposomes encapsulating siATG7, the efficient cytoplasmic delivery of siRNA was achieved. This method effectively downregulates the autophagy-related gene ATG7 and exhibits excellent antifibrotic effects both in vitro and in vivo.

Summary: This study screened efficient HIGS vectors targeting Phytophthora capsici and found that hairpin vectors with a 70 nt loop and similar to 500 bp stem showed the highest control efficacy. Further research revealed that the amount of small interfering RNAs (siRNAs) generated by the HIGS vectors was positively related to gene silencing efficiency and resistance, with NbDCL3 and NbDCL4 identified as the key effectors producing siRNAs. These findings contribute to the design of efficient HIGS vectors and shed light on the molecular mechanism of HIGS.

However, challenges in terms of pharmacokinetics and pharmacodynamics have been addressed through chemical modification and delivery systems. Several siRNA therapies have been approved for clinical use, but further advancements are needed to target organs beyond the liver and reach special sites.

Summary: Spray-induced gene silencing (SIGS) is a promising technology for disease control in plants, but it is limited by the instability of RNA. In this study, we used artificial nanovesicles (AVs) to encapsulate and protect double stranded RNA (dsRNA) against Botrytis cinerea. The results showed that AVs enabled efficient delivery and uptake of dsRNA, providing prolonged RNAi-mediated protection against B. cinerea on plant material.

Summary: The leading cause of death, cancer, necessitates the development of new effective treatment systems, such as genetic therapy using siRNA. This study explores the use of dendronized and PEGylated silver nanoparticles as potential vectors to deliver proapoptotic siRNA (siMCL-1) to target cells. Through various methods, stable siRNA and AgNP complexes were obtained and shown to efficiently deliver siRNAs to tumor cells, resulting in significant cell death due to apoptosis.

Summary: In this study, small circular interfering RNAs (sciRNAs) were synthesized and evaluated for their therapeutic potential. Circularization improved metabolic stability of the sense strand and eliminated off-target effects, resulting in potent GaINAc-sciRNAs at therapeutically relevant doses. Physicochemical studies and NMR-based structural analysis shed light on the interactions of this novel class of siRNAs with the RNAi machinery.
