A novel bacteriocin against Staphylococcus aureus from Lactobacillus paracasei isolated from Yunnan traditional fermented yogurt: Purification, antibacterial characterization, and antibiofilm activity

A novel LAB bacteriocin LSX01 isolated from traditional fermented yogurt in Yunnan, China was found to possess antibacterial and antibiofilm activity against Staphylococcus aureus, suggesting its potential in improving dairy product safety.
Staphylococcus aureus and its biofilm have emerged as a significant threat to the safety of dairy products. In recent years, lactic acid bacteria (LAB) bacteriocins have been widely acknowledged as the potential natural antibacterial substance in food biopreservation due to their excellent antibacterial effects. However, few LAB bacteriocins with antibacterial and antibiofilm activity against S. aureus have been reported in dairy products. In the present study, a novel bacteriocin LSX01 of Lactobacillus paracasei LS-6 isolated from a traditional fermented yogurt produced in Yunnan, China, was purified and characterized extensively. The LSX01 possessed a molecular weight of 967.49 Da and an AA sequence of LDQAGISYT. The minimum inhibitory concentration of LSX01 against S. aureus_45 was 16.90 mu g/mL, which was close to or lower than the previously reported bacteriocins. The LSX01 exhibited an extensive antimicrobial spectrum against both gram-positive and gram-negative bacteria. Moreover, LSX01 exhibited excellent tolerance to heat and acid-base treatments, and sensitivity to the proteolytic enzymes, such as pepsin and proteinase K. Furthermore, the treatment of S. aureus_45 planktonic cells with LSX01 significantly reduced their metabolic activity and disrupted the cell membrane integrity. Scan electron microscopy results demonstrated that LSX01 induced cytoplasmic content leakage and cell deformation. Additionally, biofilm formation of S. aureus_45 was also significantly inhibited by LSX01. Overall, the results suggested that the novel LAB bacteriocin LSX01 possessed antibacte rial activity and antibiofilm activity against S. aureus and, hence, could have potential for improving safety of dairy products.

A method for isolating highly purified and active mitochondria from insects

Summary: This study presents a method for isolating highly purified and active mitochondria from insects. The method combines differential centrifugation and discontinuous Nycodenz density gradient centrifugation and has been validated across three orders and four insect species. The study demonstrates the removal of nonmitochondrial components and the preservation of mitochondria activity/integrity. It also highlights the importance of storing the isolated mitochondria at -20 degrees C for downstream experiments.

Antibacterial activity and action target of phenyllactic acid against Staphylococcus aureus and its application in skim milk and cheese

Summary: This study demonstrates the antibacterial activity of phenyllactic acid (PLA) against Staphylococcus aureus and its potential application in dairy products. PLA inhibits the growth and biofilm formation of S. aureus, and it exerts antibacterial effects by affecting the cell metabolism, membrane integrity, intracellular content leakage, oxidative stress, and morphological deformation of S. aureus. The antibacterial effects of PLA against S. aureus cells in skim milk and cheese are maintained during storage for 21 days at 4 degrees C.

Antibacterial activity and mechanism of action of bacteriocin LFX01 against Staphylococcus aureus and Escherichia coli and its application on pork model

Summary: This study investigated the antibacterial activity and mechanism of action of bacteriocin LFX01 against two important indicator strains that cause pork contamination. The results showed that LFX01 exhibited antibacterial activity against these indicator strains by reducing cell metabolic activity, disrupting cell membrane integrity, and causing leakage of intracellular contents. Additionally, LFX01 was found to bind to the genomic DNA of the indicator strains, disrupting its structure. These findings revealed the mechanism of action of LFX01 against the indicator strains.

Subcellular-Level Mitochondrial Energy Metabolism Response in the Fat Body of the German Cockroach Fed Abamectin

Summary: This study investigated the effects of the pesticide abamectin on mitochondrial energy metabolism using metabolomic analysis of isolated mitochondria from German cockroaches. The results showed abnormal mitochondrial morphology and changes in key enzyme activities involved in energy metabolism under abamectin stress. Metabolomic analysis revealed a set of metabolites and pathways related to mitochondrial energy metabolism. This study provides valuable insights into mitochondrial toxicology at a subcellular level under environmental toxicants.
