Water-Soluble Iridium Photoredox Catalyst for the Trifluoromethylation of Biomolecule Substrates in Phosphate Buffered Saline Solvent

Summary: The development of a water-soluble iridium catalyst enables the trifluoromethylation of polar small molecules and peptides in DMSO solution or aqueous media, providing up to 60% CF3- peptide. This reaction was optimized in a microtiter plate format under ambient air, using commercial Langlois reagent as a CF3 radical source and blue LEDs for excitation.

Silver Benzoate Facilitates the Copper-Catalyzed C-N Coupling of Iodoazoles with Aromatic Nitrogen Heterocycles

Summary: Traditional C-N coupling methods for the reaction of iodo-oxazole with 2-pyridinone was found to be low yielding, but yield can be improved by adding silver benzoate additives, CuI catalysts, and specific ligands. This method is applicable for the coupling of various iodo-heterocycles with 2-pyridinone, as well as for coupling NH-containing heterocycles with 4-iodooxazole.

Summary: Parkinson's disease is a neurodegenerative disorder with no effective treatments currently available. However, research suggests that Renin-Angiotensin-Aldosterone System (RAAS) inhibitors may be beneficial in treating and delaying the progression of the disease by restoring mitochondrial function to protect dopamine-producing neurons. Further studies and improvements in these drugs are needed to explore and capitalize on their potential benefits.

Different α-synuclein prion strains cause dementia with Lewy bodies and multiple system atrophy

Summary: Alpha-synuclein protein, with different conformations, is associated with neurodegenerative diseases including multiple system atrophy, dementia with Lewy bodies, and Parkinson's disease. Through experimental studies using transgenic mice and cultured cells, different strains of alpha-synuclein prions are found to be responsible for these diseases.

Stacked binding of a PET ligand to Alzheimer's tau paired helical filaments

Summary: The accumulation of tau protein aggregates in the brain is a characteristic feature of Alzheimer's disease and other neurodegenerative tauopathies, and understanding the mode of small-molecule binding to these aggregates is critical for diagnosis and treatment. The authors used cryo-electron microscopy to determine the structure of AD patient-derived tau filaments bound to the PET ligand GTP-1, revealing a stacked binding motif that may aid in the design of diagnostics and therapeutics. This study provides insight into the development of compounds targeting different amyloid folds found in neurodegenerative diseases.
