Summary: The study focuses on the kinetics of a water-soluble host-guest system based on cucurbit[8]uril and two DTE photoswitches, demonstrating a method to control the binding kinetics in supramolecular systems using external stimuli. By modifying the structure of the DTE guest molecule, it is possible to significantly slow down the formation/dissociation rates of pseudorotaxane structures, leading to the emergence of a competitive metastable product with important impact on the overall binding kinetics. Additionally, the dissociation kinetics of the host-guest system can be controlled by approximately 100-fold by switching between closed and open DTE isomers.

Summary: The interactions between neutral/protonated mitoxantrone and cucurbit[n]uril (n = 7, 8) host systems were analyzed using density functional theory calculations. It was found that stable inclusion complexes can form between doubly and quadruply charged mitoxantrone molecules and CB[7] or CB[8] in a water solvent. Both CB[7] and CB[8] show properties that are useful for drug delivery.

Summary: In this study, four supramolecular organic framework microcrystals were prepared as efficient adsorbents for bilirubin removal in acute liver failure patients. The adsorption kinetics of these microcrystals were fast and the adsorption capacity was high. Specifically, one of the microcrystals, SOF-3, was further processed into a composite with poly(ether sulfone) and showed effective bilirubin adsorption in a dynamic hemoperfusion process. This work provides a new approach for the development of novel bilirubin adsorbents for hemoperfusion therapy.

Summary: Paracyclophane has been understudied compared to other macrocyclic compounds, but through modification of pillar[5]arene, new pentagonal macrocycles (pseudo[n]-pillar[5]arenes) were obtained. These macrocycles showed the ability to form complexes with various guests in a 1:1 ratio. The binding constants decreased with the decrease of substituted phenylene segments. Notably, the conformation of P[n]P[5]s could adjust effectively when binding with succinonitrile in the solid state.

These supramolecular probes are capable of detecting the acidic environment in endosomal compartments in live cells.

Summary: In the past two decades, photodynamic therapy (PDT) has been effective for cancer treatment. However, the residue of photodynamic agents (PDAs) causes long-term skin phototoxicity. This study uses NpBoxes to bind to porphyrin-based PDAs and reduce their content in skin tissues, alleviating posttreatment phototoxicity. The compound 2,6-NpBox can efficiently suppress photosensitivity and reactive oxygen species generation without affecting PDT efficacy.

Summary: Acyclic cucurbit[n]uril-based nanosponges were prepared using supramolecular vesicle-templated cross-linking and successfully loaded with the photodynamic therapeutic drug temoporfin. The nanosponges significantly enhanced the PDT bioactivity of temoporfin against cancer cells and were found to be non-toxic and biocompatible in vitro and in vivo.
