Supramolecular organic frameworks improve the safety of clinically used porphyrin photodynamic agents and maintain their antitumor efficacy

In this study, three-dimensional diamondoid supramolecular organic frameworks (SOFs) were found to effectively suppress the skin phototoxicity of clinically used photodynamic agents (PDAs). The results showed that SOFs have strong interactions with PDAs, can adsorb PDAs at a micromolar concentration, and have high biocompatibility. In animal experiments, SOFs were able to suppress sunlight-induced skin phototoxicity without affecting the efficacy of photodynamic therapy (PDT).
Despite that photodynamic therapy (PDT) has been applied for the treatment of cancer and skin diseases for more than two decades, all clinically used photodynamic agents (PDAs) suffer the drawback of skin phototoxicity of PDAs, which requires patients to avoid exposure to natural light for weeks after treatment, but has so far lacked effective suppression methods. Here, we report that three-dimensional diamondoid supramolecular organic frameworks (SOFs), that possess well-defined 2.1-nm porosity, can be used to suppress the skin phototoxicity of Photofrin, HiPorfin and Talaporfin, three porphyrin-based PDAs which clinically receive the most wide applications by injecting SOF after PDT, via an adsorption and retention mechanism. Fluorescence and dynamic light scattering experiments confirm that the SOFs have strong interaction with PDAs, and can adsorb PDAs at a micromolar concentration, whereas dialysis experiments support that the adsorption leads to an important retention effect. In vitro and in vivo experiments reveal that SOFs have high biocompatibility. Studies with healthy and tumor-bearing mouse models demonstrate that, when the PDAs are administrated at a dose comparable with the clinical one, SOF can remarkably suppress sunlight-induced skin phototoxicity, whereas the PDT efficacy of mice treated with SOF post-PDT is maintained. This work provides an efficient strategy for the improvement of the safety of clinically used PDAs.

Supramolecular Organic Frameworks as Adsorbents for Efficient Removal of Excess Bilirubin in Hemoperfusion

Summary: In this study, four supramolecular organic framework microcrystals were prepared as efficient adsorbents for bilirubin removal in acute liver failure patients. The adsorption kinetics of these microcrystals were fast and the adsorption capacity was high. Specifically, one of the microcrystals, SOF-3, was further processed into a composite with poly(ether sulfone) and showed effective bilirubin adsorption in a dynamic hemoperfusion process. This work provides a new approach for the development of novel bilirubin adsorbents for hemoperfusion therapy.

Pseudo[n]-pillar[5]arenes: Synthesis, Structures, and Host-Guest Binding Properties

Summary: Paracyclophane has been understudied compared to other macrocyclic compounds, but through modification of pillar[5]arene, new pentagonal macrocycles (pseudo[n]-pillar[5]arenes) were obtained. These macrocycles showed the ability to form complexes with various guests in a 1:1 ratio. The binding constants decreased with the decrease of substituted phenylene segments. Notably, the conformation of P[n]P[5]s could adjust effectively when binding with succinonitrile in the solid state.

pH-sensing supramolecular fluorescent probes discovered by library screening

Summary: A new design concept for pH-sensing supramolecular fluorescent probes is presented. These probes are designed based on pro-guests, which degrade under acidic conditions and displace encapsulated dyes to enhance fluorescence intensity. A library of potential probes is generated and screened for effective detection of acidic pH in solution. These supramolecular probes are capable of detecting the acidic environment in endosomal compartments in live cells.

Host-guest binding of tetracationic cyclophanes to photodynamic agents inhibits posttreatment phototoxicity and maintains antitumour efficacy

Summary: In the past two decades, photodynamic therapy (PDT) has been effective for cancer treatment. However, the residue of photodynamic agents (PDAs) causes long-term skin phototoxicity. This study uses NpBoxes to bind to porphyrin-based PDAs and reduce their content in skin tissues, alleviating posttreatment phototoxicity. The compound 2,6-NpBox can efficiently suppress photosensitivity and reactive oxygen species generation without affecting PDT efficacy.

Acyclic cucurbit[n]uril-based nanosponges significantly enhance the photodynamic therapeutic efficacy of temoporfin in vitro and in vivo

Summary: Acyclic cucurbit[n]uril-based nanosponges were prepared using supramolecular vesicle-templated cross-linking and successfully loaded with the photodynamic therapeutic drug temoporfin. The nanosponges significantly enhanced the PDT bioactivity of temoporfin against cancer cells and were found to be non-toxic and biocompatible in vitro and in vivo.

Summary: Water-soluble flexible organic frameworks (FOFs) were designed and prepared to sequester and prevent the escape of neuromuscular blocking agents (NMBAs). FOFs, with different pore sizes, were shown to bind NMBAs in water, exhibiting a multivalency interaction. Animal studies demonstrated that FOFs effectively reversed neuromuscular block induced by NMBAs.
