Sex as an important factor in nanomedicine

The role and impact of sex in nanomedicine are crucial, including differences at the cellular and molecular levels. Researchers should pay more attention to the role of sex physiology in nanomedicine to design and develop safer and more efficient sex-specific nanomedicine products.
Nanomedicine has demonstrated substantial potential to improve the quality and efficacy of healthcare systems. Although the promise of nanomedicine to transform conventional medicine is evident, significant numbers of therapeutic nanomedicine products have failed in clinical trials. Most studies in nanomedicine have overlooked several important factors, including the significance of sex differences at various physiological levels. This report attempts to highlight the importance of sex in nanomedicine at cellular and molecular level. A more thorough consideration of sex physiology, among other critical variations (e.g., health status of individuals), would enable researchers to design and develop safer and more-efficient sex-specific diagnostic and therapeutic nanomedicine products. Given the early failing of therapeutic nanomedicine in the clinic the importance of understanding bio-nano interactions and biosystem parameters has gained much attention. Here, the authors review the work to date that looks at the impact of sex on the applications and outcomes of nanomedicine.

Recent advances in nanoscale targeted therapy of HER2-positive breast cancer

Summary: Breast cancer is the second major cause of death among women worldwide, with a high mortality rate. The unique tumor microenvironment created by breast cancer cells allows them to evade immune surveillance, making conventional chemotherapy and immunotherapy ineffective. Overexpressed tumor markers in breast cancer cells can be targeted for detection and therapy, with HER2 being a clinically valid molecular target.

Advanced biological and engineering methods are utilized to enhance and customize tissue engineered scaffolds, while tunable drug delivery mechanisms are incorporated to address clinical challenges. The review covers the mechanisms and recent developments in drug delivery vehicles, loading strategies, and monitoring/release kinetics of embedded drugs in engineered scaffolds, as well as the trend of using stimuli driven biopolymer scaffolds for precise spatiotemporal control of drug release.

This review summarizes the effects of environmental chemical exposure on cellular redox signaling, ferroptosis, and cell death, providing critical information to mitigate the impact of ferroptosis on human health.

Tumor acidic environment directs nanoparticle impacts on cancer cells

Summary: This study investigates the biological impacts of different nanoparticles on cancer cells at different pH levels. The researchers found that a slight change in the pH of the cancer cell microenvironment significantly affects the cellular uptake efficacy and toxicity mechanism of the nanoparticles. The nanoparticles exhibited more toxic effects against cancer cells at physiological pH compared to acidic pH, and they also affected different phases of the cell cycle at different pH levels. Additionally, cancer cells expressed higher levels of interleukins involved in cancer cell resistance at acidic pH. The findings highlight the importance of considering the extracellular pH of cancer cells in future cancer nanomedicine research.

An Overview of Nanoparticle Protein Corona Literature

Summary: The protein corona is formed spontaneously on nanoparticle surfaces in biological systems/fluids. Characterizing the protein corona is crucial for the development of safe and efficient diagnostic and therapeutic nanomedicine products. This review analyzed 470 papers from 2000 to 2021, covering 1702 nanoparticle systems, and revealed key findings regarding the types of nanoparticles studied, the underrepresentation of lipid-based NPs in corona research, the use of new methods to improve reliability, the focus on personalized medicine, and the importance of careful characterization of nanoparticles after corona formation.

Identification of the Proteins Determining the Blood Circulation Time of Nanoparticles

Summary: The therapeutic efficacy and side effects of nanoparticles (NPs) depend on their circulation time in the body. The adsorbed coronaproteins on NPs determine their plasma half-lives, and it is important to identify the proteins that influence their circulation time. This study analyzed the in vivo circulation time and protein composition of superparamagnetic iron oxide nanoparticles (SPIONs) with different surface charges/chemistries. SPIONs with neutral and positive charges had the longest and shortest circulation times, respectively. The composition of the corona-coated NPs also affected their circulation time, with certain proteins playing a role in determining their systemic circulation time.

Mass Spectrometry, Structural Analysis, and Anti-Inflammatory Properties of Photo-Cross-Linked Human Albumin Hydrogels

Summary: Albumin-based hydrogels, especially the non-immunogenic photocurable version described in the study, demonstrate excellent biocompatibility and anti-inflammatory properties, making them promising materials for biomedical applications. The modifications and cross-linking of methacrylated HSA were found to reduce macrophage adhesion and inflammatory pathways, with potential for use in passivation coatings and other biomedical applications. In vitro and Ex-Ovo assay results confirmed the biocompatibility and slight angiogenesis-modulating effects of the photocurable albumin hydrogels.

Conformation- and phosphorylation-dependent electron tunnelling across self-assembled monolayers of tau peptides

Summary: We investigated the charge transport characteristics of self-assembled monolayers (SAMs) of short tau peptides using electron tunneling rates and quantum mechanical simulation. Our findings showed that the conformation and phosphorylation of short peptides can significantly affect their electron tunneling current and energy levels.
