Exhaled patient derived aerosol dispersion during awake tracheal intubation with concurrent high flow nasal therapy

Summary: Awake Tracheal Intubation (ATI) is a potential risk for healthcare workers due to the generation of aerosol particles. This study used optical particle sizers to observe aerosol distribution during a simulated ATI with high-flow nasal oxygen therapy. The highest concentration of patient-derived aerosol particles was within 0.5-1.0 m of the subject, while particles > 5 μm spread radially throughout the operating room. It is recommended that full airborne protective equipment be worn by all staff during ATI on patients with suspected viral respiratory infections.

Aerosolized Pulmonary Delivery of mRNA Constructs Attenuates Severity of Escherichia coli Pneumonia in the Rat

Summary: ARDS is a rapid-onset inflammatory lung disease without effective specific therapy, for which pneumonia is a common pathogenic etiology. Previous studies have shown that prophylactic delivery of mRNA coding for NF-kappa B inhibitor alpha super-repressor (I kappa B alpha-SR) or extracellular superoxide dismutase 3 (SOD3) by viral vector can help reduce the severity of pneumonia. This study demonstrates that nebulized mRNA therapeutics, complexed with cationic lipid and delivered through a vibrating mesh nebulizer, have potential as a promising approach for ARDS therapy. The mRNA treatments resulted in rapid expression of proteins and observable improvement in pneumonia symptoms.

Summary: Oxidative stress, primarily caused by reactive oxygen species (ROS) produced by impaired mitochondria, is a major feature of COPD. While antioxidants have shown potential in mitigating oxidative damage, currently available agents are not widely used. Mitochondria-targeted antioxidants (MTAs) offer a more precise approach to reducing ROS and have demonstrated greater protective effects in COPD, making them promising therapeutic agents. However, challenges and future directions for the use of MTAs in COPD treatment need to be discussed.

This review discusses the historical background and development of SLNs in nanotechnology. It also focuses on the composition, fabrication techniques, mechanism, and advantages of SLNs. The review highlights the biopharmaceutical aspects and challenges associated with the clinical translation of SLNs for improving the bioavailability, stability, and solubility of lipophilic and gastrointestinal labile drugs used in treating chronic diseases.

Cigarette smoking induces lung cancer tumorigenesis via upregulation of the WNT/β-catenin signaling pathway

Summary: Lung cancer has the highest mortality rate worldwide, and cigarette smoking is a major etiological factor. This study found that cigarette smoke extract treatment can upregulate WNT/ss-β-catenin signaling and induce tumorigenesis in healthy human bronchial epithelial cells. Furthermore, extracellular vesicles obtained from cigarette smoke exposed cells were found to induce migration and upregulate oncology proteins in recipient cells.

Summary: Excessive inflammation-associated coagulation is a common feature in infectious diseases such as bacterial sepsis and COVID-19. Type I interferon signaling has been found to play a crucial role in the release of tissue factor (TF) from macrophages, an important initiator of coagulation. The drugs DMF and 4-OI have been identified as anticoagulants that inhibit TF-mediated coagulopathy by suppressing macrophage pyroptosis and subsequent TF release.
