The Cre/Lox system is a powerful tool for gene studies, allowing for loss-of-function and gain-of-function experiments. However, Cre transgenic lines may have off-target activity, emphasizing the importance of selecting optimal Cre lines for research purposes.
The Cre/Lox system is a powerful tool in the biologist's toolbox, allowing loss-of-function and gain-of-function studies, as well as lineage tracing, through gene recombination in a tissue-specific and inducible manner. Evidence indicates, however, that Cre transgenic lines have a far more nuanced and broader pattern of Cre activity than initially thought, exhibiting off-target activity in tissues/cells other than the ones they were originally designed to target. With the goal of facilitating the comparison and selection of optimal Cre lines to be used for the study of gene function, we have summarized in a single manuscript the major sites and timing of Cre activity of the main Cre lines available to target bone mesenchymal stem cells, chondrocytes, osteoblasts, osteocytes, tenocytes, and osteoclasts, along with their reported sites of off-target Cre activity. We also discuss characteristics, advantages, and limitations of these Cre lines for users to avoid common risks related to overinterpretation or misinterpretation based on the assumption of strict cell-type specificity or unaccounted effect of the Cre transgene or Cre inducers. Mechanisms Supporting the Use of Beta-Blockers for the Management of Breast Cancer Bone Metastasis

Maria-Bernadette Madel, Florent Elefteriou

Summary: Bone is a common site for breast cancer metastasis, impacting patient survival and quality of life, with the interaction between sympathetic nerves and bone cells influencing this process. Stimulation of beta-adrenergic receptors in stromal bone cells promotes the establishment of disseminated cancer cells in the skeleton.

Summary: Mechanical loading stimulates bone formation, and the Wnt signaling pathway plays a crucial role in the skeletal response to loading. Specifically, Wnt1 mediates the mechano-adaptive response to loading.

Summary: This study assessed the ability of matrix-sensitive techniques to differentiate between NF1 mice and wild-type mice. The results showed that low bound water and cortical volumetric bone mineral density were the most accurate characteristics for distinguishing between the two genotypes in femurs, while cyclic reference point indentation tests' average unloading slope was the most sensitive in differentiating tibias. These findings provide guidance for the clinical trials of NF1.

Summary: Major achievements in bone research have relied on animal models and in vitro systems. However, the ethical debate surrounding animal experimentation and advancements in in vitro and in silico techniques have challenged their use. This review provides an overview of the strengths and limitations of animal models, as well as in vitro and in silico technologies, in skeletal research. The authors propose that a combination of the right animal model and state-of-the-art technology is essential for advancing bone research.
