Summary: Leukoencephalopathy with calcifications and cysts (LCC) is a neurological syndrome associated with pathogenic variants in SNORD118. Neuropathological findings in a genetically confirmed LCC individual revealed diffuse destructive process due to a vasculopathy with secondary ischemic lesions and mineralization. Treatment with Bevacizumab may offer clinical and radiological benefit for LCC patients, especially in cases where rapid diagnosis and trial of this treatment modality is considered appropriate.

Summary: Aicardi-Goutières syndrome (AGS) is a monogenic type-I interferonopathy that results in neurologic injury and is associated with liver inflammation. All genotypes of AGS are associated with transient elevations of liver enzymes and the presence of liver-associated autoantibodies.

Summary: This study examined the relationship between body mass index (BMI) and disability in children with Charcot-Marie-Tooth disease (CMT). The results showed that compared to healthy children, children with CMT had a higher proportion of being severely underweight, underweight, and obese, while having a lower proportion of being at a healthy weight. Furthermore, being underweight or obese was associated with greater disability in children with CMT compared to those of a healthy weight.

Summary: This study aimed to evaluate the impact of body mass index (BMI) on disease progression in children with Charcot-Marie-Tooth disease (CMT) over a period of 2 years. The results showed that children with CMT who were underweight or obese exhibited greater disability. It suggests that interventions to maintain or improve BMI towards a healthy weight may reduce disability in children with CMT.

Summary: This study identified rare NMNAT2 gene variants associated with a neuropathy syndrome and demonstrated the importance of SARM1-related neuroimmune mechanisms in disease pathogenesis. Macrophage depletion therapy was shown to block and reverse neuropathic phenotypes in mice with NMNAT2 mutations.
