Chemokines and relapses in childhood acute lymphoblastic leukemia: A role in migration and in resistance to antileukemic drugs

We studied whether chemokines may have a role in relapses in childhood acute lymphoblastic leukemia (ALL). We compared the levels of chemokine receptors in marrow samples from 82 children with ALL at diagnosis versus 15 at relapses, and quantified the levels of chemokines in central system fluid (CSF) samples. The functional role of specific chemokines was studied in vitro and in vivo. The expression of some chemokine receptors was upregulated upon leukemic relapse, both in B- and in T-ALL, and in cases of medullary and extramedullary involvement. CXCL10 induced chemotaxis in leukemic cell lines and in primary leukemic cells, depending upon the levels of CXCR3 expression. CXCL10 specifically diminished chemotherapy-induced apoptosis on ALL cells expressing CXCR3, partially inhibiting caspase activation and maintaining the levels of the antiapoptotic protein Bcl-2. Finally, immunodeficient mice engrafted with CXCR3-expressing human leukemic cells showed decreased infiltration of marrow, spleen, and CNS after receiving a OCCR3-antagonist molecule. CXCR3 signaling in ALL may have a dual function: chemotactic for the localisation of leukemic blasts in specific niches, and it may also confer resistance to chemotherapy, enhancing the chances for relapses. Dual-Target CAR-Ts with On- and Off-Tumour Activity May Override Immune Suppression in Solid Cancers: A Mathematical Proof of Concept

Summary: CAR-T cell-based therapies have shown success in hematological malignancies but have been limited in solid tumors due to the immunosuppressive tumor microenvironment. A mathematical model in this paper suggests that targeting solid tumor antigens with large numbers of CAR-T cells could overcome this immunosuppression, potentially leading to successful treatments. Computer simulations indicate that the strategy of utilizing CAR-T cells targeting multiple antigens may lead to the expansion of a strong army of CAR-T cells in solid tumors. This proposed approach could provide a promising avenue for overcoming the immune suppression and achieving successful CAR-T cell therapies against solid tumors.

Bayesian Inference in Y-Linked Two-Sex Branching Processes with Mutations: ABC Approach

Summary: The study analyzes the evolution of Y-linked allele carriers and mutations using a Y-linked two-sex branching process model, estimating main parameters through Bayesian inference. By observing total number of females, males, males of each genotype in last generation, and introducing information on mutated males in last generation, the study approximates the posterior distributions of model parameters using Approximate Bayesian Computation (ABC). Illustrations and discussion on accuracy of the procedure based on simulated examples are provided.

Phenolic and quinone methide nor-triterpenes as selective NLRP3 inflammasome inhibitors

Summary: This study evaluated the potential anti-inflammatory activity of phenolic and quinonemethide nor-triterpenes isolated from Maytenus retusa and their derivatives through inhibition of the NLRP3 inflammasome. Triterpenes 6 and 14 showed the highest anti-inflammatory activity by significantly reducing caspase-1 activity, IL-1 beta secretion, and pyroptosis. Further characterization confirmed their selective inhibition of NLRP3 inflammasome without affecting AIM2 or NLRC4 inflammasome activation.

Synthesis of Quinoline and Dihydroquinoline Embelin Derivatives as Cardioprotective Agents

Summary: A set of new dihydroquinoline embelin derivatives were synthesized from the reaction of natural benzoquinone embelin with anilines and aromatic aldehydes in the presence of AgOTf. The resulting compounds showed potential as cardioprotective agents, attenuating the cardiotoxicity effect of doxorubicin on oxidative stress and apoptosis. The best activities were achieved with quinoline-embelin derivatives having a 4-nitrophenyl group attached at the pyridine ring.

Secondary salvage use of cord blood transplantation may still be useful for patients who have failed after other transplantation.

Bach1 inhibitor HPP-D mediates ?-globin gene activation in sickle erythroid progenitors

Summary: This study evaluated the ability of a novel Bach1 inhibitor, HPP-D, to induce HbF in KU812 cells and primary sickle erythroid progenitors. The results showed that HPP-D increased HbF levels and decreased Bach1 protein levels in both cell types. Furthermore, it enhanced NRF2 binding to the gamma-globin promoter antioxidant response elements, leading to an open chromatin configuration and increased gamma-globin transcription.
