Myeloid-derived suppressor cells (MDSCs) are a heterogeneous group of myeloid progenitor and precursor cells at different stages of differentiation, which play an important role in tumor immunosuppression. Glioma is the most common and deadliest primary malignant tumor of the brain, and ample evidence supports key contributions of MDSCs to the immunosuppressive tumor microenvironment, which is a key factor stimulating glioma progression. In this review, we summarize the source and characterization of MDSCs, discuss their immunosuppressive functions, and current approaches that target MDSCs for tumor control. Overall, the review provides insights into the roles of MDSC immunosuppression in the glioma microenvironment and suggests that MDSC control is a powerful cellular therapeutic target for currently incurable glioma tumors.

Cross-platform analysis reveals cellular and molecular landscape of glioblastoma invasion

Ann T. Chen, Yang Xiao, Xiangjun Tang, Mehdi Baqri, Xingchun Gao, Melanie Reschke, Wendy C. Sheu, Gretchen Long, Yu Zhou, Gang Deng, Shenqi Zhang, Yanxiang Deng, Zhiliang Bai, Dongjoo Kim, Anita Huttner, Russell Kunes, Murat Gunel, Jennifer Moliterno, W. Mark Saltzman, Rong Fan, Jiangbing Zhou

Summary: This study investigates the invasion of glioblastoma (GBM) through the analysis of histological and single-cell RNA sequencing (scRNA-seq) data. The research reveals the cellular and molecular characteristics of invasive GBM, including lower cell density and proliferation rate compared to non-invasive GBM. Additionally, the study identifies the genes CRYAB and CD44 as important regulators of tumor invasion and highlights their relationship in invasive GBM.

Remodeling liver microenvironment by L-arginine loaded hollow polydopamine nanoparticles for liver cirrhosis treatment

Yeying Wang, Yang Liu, Yi Liu, Jie Zhong, Jing Wang, Lei Sun, Lei Yu, Yiting Wang, Qinghua Li, Weilin Jin, Zhiqiang Yan

Summary: Liver cirrhosis is a liver disease with a high mortality rate worldwide. Antifibrotic drugs are commonly used clinically, but there are still many challenges. Excessive reactive oxygen species (ROS) in the liver lesions microenvironment is an important factor leading to the development of liver cirrhosis. Therefore, a nanomedicine-mediated antioxidant therapy can remodel the liver microenvironment and reverse the process of cirrhosis.

CBL0137 activates ROS/BAX signaling to promote caspase-3/GSDME-dependent pyroptosis in ovarian cancer cells

Chang Yang, Zhi-Qiang Wang, Zhao-Cong Zhang, Ge Lou, Wei-Lin Jin

Summary: The study found that CBL0137 induces apoptosis and pyroptosis in ovarian cancer cells by activating the reactive oxygen species pathway and regulating the BAX protein. In vivo experiments also demonstrated its anti-tumor effects on ovarian cancer cells. These findings suggest that CBL0137 could be a promising therapeutic agent for ovarian cancer.

Adipocyte-derived extracellular vesicles: bridging the communications between obesity and tumor microenvironment

Chuan Zhou, Yu-Qian Huang, Ming-Xu Da, Wei-Lin Jin, Feng-Hai Zhou

Summary: By 2035, more than 4 billion people might be affected by obesity and being overweight. Adipocyte-derived Extracellular Vesicles (ADEVs/ADEV-singular) play a crucial role in the communication between the tumor microenvironment and obesity, serving as a key mechanism of tumor progression. Obesity leads to hypertrophic and hyperplastic adipose tissue, causing insulin resistance and altering the energy supply to tumor cells, as well as promoting the production of pro-inflammatory adipokines. ADEVs are closely associated with cancer hallmarks and have the potential to be used as biomarkers and antitumor therapy. It is important to address the significant challenges and advances in ADEVs research and clinical applications.

Collagen code in tumor microenvironment: Functions, molecular mechanisms, and therapeutic implications

Qian Zhang, Zi-Yi An, Wen Jiang, Wei-Lin Jin, Xin-Yang He

Summary: “Collagen, a major component of the extracellular matrix (ECM), plays a crucial role in tumor progression within the tumor microenvironment (TME). Recent studies have shown that collagen deposition in the TME affects tumor progression and interacts with tumor cells in complex ways. These findings highlight the potential of collagen as a target for cancer therapy.”
