Chitosan/Poly(2-ethyl-2-oxazoline) Films with Ciprofloxacin for Application in Vaginal Drug Delivery

Chitosan (CHI) and chitosan/poly(2-ethyl-2-oxazoline) (CHI/POZ)-based films were prepared by casting from aqueous solutions of polymer blends with different compositions. Ciprofloxacin was used as a model drug in these formulations. The weight, thickness, folding endurance and transparency of blend films were measured and characterised. All films had a uniform thickness (0.06 +/- 0.01 mm) and exhibited sufficient flexibility. The surface pHs of films ranged from 3.76 +/- 0.49 to 4.14 +/- 0.32, which is within the pH range suitable for vaginal applications. The cumulative release of the drug from the films in experiments in vitro was found to be 42 +/- 2% and 56 +/- 1% for pure CHI and CHI/POZ (40:60) films, respectively. Drug-free chitosan/poly(2-ethyl-2-oxazoline) films showed weak antimicrobial activity against Escherichia coli. Drug-loaded CHI and CHI/POZ films showed good antimicrobial properties against both Gram-positive Staphylococcus aureus and Gram-negative bacteria Escherichia coli. Mucoadhesive properties of these films with respect to freshly excised sheep vaginal mucosa were evaluated using a tensile method. It was established that all films were mucoadhesive, but an increase in POZ content in the blend resulted in a gradual reduction of their ability to stick to vaginal mucosa. These films could potentially find applications in vaginal drug delivery.

Nanotechnology-based drug delivery systems have been investigated as effective alternatives to conventional ocular delivery systems for treating anterior segment diseases. This review discusses different nanotechnology-based delivery systems and their prospects in clinical use.

Synthesis of Methacryloylated Hydroxyethylcellulose and Development of Mucoadhesive Wafers for Buccal Drug Delivery

Summary: Non-ionic hydroxyethylcellulose (HEC) was chemically modified with glycidyl methacrylate to enhance its mucoadhesive properties. The modification was confirmed by H-1 NMR and FTIR spectroscopy, and quantification of methacryloyl groups was done using HPLC. Toxicity studies showed no toxicological effects of the modified HEC. Mucoadhesive wafers developed from the modified HEC exhibited significantly better mucoadhesive properties compared to unmodified HEC.

Cataleptogenic Effect of Haloperidol Formulated in Water-Soluble Calixarene-Based Nanoparticles

Summary: A water-soluble form of haloperidol was obtained by coaggregation with calix[4]resorcinol to form vesicular nanoparticles. The nanoparticles showed mucoadhesive and thermosensitive properties, making them potentially effective drug delivery systems. In pharmacological studies, haloperidol formulated with calix[4]resorcinol exhibited a cataleptogenic effect in rats both when administered intranasally and intraperitoneally.

Biodegradable Hydrogels Based on Chitosan and Pectin for Cisplatin Delivery

Summary: Preparation of stable hydrogels using charge-complementary polyelectrolyte chains offers practical advantages over the use of organic crosslinking agents. This study utilizes natural polyelectrolytes-chitosan and pectin-with their biocompatibility and biodegradability. Different molecular weights of pectin are used to prepare hydrogels with varying rheological characteristics and swelling kinetics. These polyelectrolyte hydrogels can release the cytostatic drug cisplatin over an extended period, potentially improving the effectiveness of cancer treatment.

Hydroxyethyl cellulose functionalised with maleimide groups as a new excipient with enhanced mucoadhesive properties

Summary: Hydroxyethylcellulose (HEC) is a water-soluble polymer with poor mucoadhesive properties. This study explores the modification of HEC by conjugation with maleimide groups to improve its mucoadhesive properties. The modified HEC exhibits superior mucoadhesive properties compared to unmodified HEC, forming a strong bond with mucin. The safety and efficacy of the newly synthesized polymer derivatives were also evaluated.

Preparation and Characterization of a Preformed Polyampholyte Particle Gel Composite for Conformance Control in Oil Recovery

Summary: Preformed particle gels (PPGs) based on acrylamide, (3-acrylamidopropyl) trimethylammonium chloride, and 2-acrylamido-2-methyl-1-propanesulfonic acid sodium salt were synthesized. The swelling behavior of PPGs depending on various factors was studied, and it was found that adding clay mineral improved the mechanical properties and allowed control over the Young's modulus and swelling degree of the samples.

In Situ Entrapment of Catalase within Macroporous Cryogel Matrix for Ethanol Oxidation: Flow-through Mode versus Batch Reactor

Summary: This study investigated the biocatalytic oxidation of ethanol to acetaldehyde using a catalase entrapped within a monolithic polyampholyte cryogel as the catalyst. The polyampholyte cryogels with varying amounts of catalase were synthesized in situ by cryo-polymerization of anionic monomer AMPS and cationic monomer APTAC. Under optimal conditions (temperature 10-20°C, pH 6.9-7.1, ethanol to hydrogen peroxide volume ratio 50:50), high yields of acetaldehyde from ethanol were achieved.

Mucoadhesive pickering nanoemulsions via dynamic covalent chemistry

Summary: Spherical aldehyde-functionalized nanoparticles were prepared through aqueous emulsion polymerization, and when combined with oil droplets, formed nanoemulsions. The aldehyde-functionalized nanoparticles showed stronger adhesion to sheep nasal mucosal tissue due to the formation of imine bonds.

Pickering emulsions stabilised with oligoglycine-functionalised nanodiamond as a model system for ocular drug delivery applications

Summary: This study evaluated the use of nanoparticles-stabilised Pickering emulsions for the confinement of organic components in ocular drug delivery. The results showed that these emulsions had good stability and mucoadhesive properties, and were non-toxic to tissues. These emulsions have distinct advantages for ocular drug delivery and could serve as a reference for future formulation design for drug delivery.

Dynamic light scattering and transmission electron microscopy in drug delivery: a roadmap for correct characterization of nanoparticles and interpretation of results

Summary: This focus article provides a detailed analysis of transmission electron microscopy (TEM) and dynamic light scattering (DLS) as two common methods for studying nanoparticle sizes in pharmaceutics and drug delivery. The article highlights the importance of controlling nanoparticle size and shape in biomedical systems and discusses the discrepancies in size measurements obtained from TEM and DLS. The authors convincingly demonstrate that properly conducted experimental procedures and careful analysis of results can eliminate or reduce the mismatch between DLS and TEM data.

Aldehyde-functional thermoresponsive diblock copolymer worm gels exhibit strong mucoadhesion

Summary: A series of thermoresponsive diblock copolymer worm gels with adjustable aldehyde content were prepared using RAFT aqueous dispersion polymerization. The aldehyde-functional worm gels showed comparable mucoadhesion performance to chitosan, a widely regarded positive control. The optimum degree of aldehyde functionality was found to be around 30%.
