Modulation of the Gut Microbiota during High-Dose Glycerol Monolaurate-Mediated Amelioration of Obesity in Mice Fed a High-Fat Diet

Obesity and associated metabolic disorders are worldwide public health issues. The gut microbiota plays a key role in the pathophysiology of diet-induced obesity. Glycerol monolaurate (GML) is a widely consumed food emulsifier with antibacterial properties. Here, we explore the anti-obesity effect of GML (1,600 mg/kg of body weight) in high-fat diet (HFD)-fed mice. HFD-fed mice were treated with 1,600 mg/kg GML. Integrated microbiome, metabolome, and transcriptome analyses were used to systematically investigate the metabolic effects of GML, and antibiotic treatment was used to assess the effects of GML on the gut microbiota. Our data indicated that GML significantly reduced body weight and visceral fat deposition, improved hyperlipidemia and hepatic lipid metabolism, and ameliorated glucose homeostasis and inflammation in HFD-fed mice. Importantly, GML modulated HFDinduced gut microbiota dysbiosis and selectively increased the abundance of Bifidobacterium pseudolongurn. Antibiotic treatment abolished all the GML-mediated metabolic improvements. A multiomics (microbiome, metabolome, and transcriptome) association study showed that GML significantly modulated glycerophospholipid metabolism, and the abundance of Bifidobacterium pseudolongum strongly correlated with the metabolites and genes that participated in glycerophospholipid metabolism. Our results indicated that GML may be provided for obesity prevention by targeting the gut microbiota and regulating glycerophospholipid metabolism.

Summary: This study aimed to explore novel antidiabetic strains from Lactobacillus plantarum and evaluated their potential through in vitro and in vivo models. Lactobacillus plantarum strain YJ7 showed high adhesion rates and antioxidant abilities, while strain N1 exhibited good inhibitory activity against α-glucosidase. In vivo studies demonstrated that YJ7 supplementation prevented insulin resistance and inflammation in high-fat diet fed mice.

Microbial diversity and chemical property analyses of sufu products with different producing regions and dressing flavors

Summary: This study found that many symbiont fungi of soybean were present in sufu samples, while lactic acid bacteria were abundant in all samples. According to the principal coordinate analysis and hierarchical clustering analysis, the bacterial communities of sufu samples could generally be clustered into the LAB clade and Bacillus clade. Furthermore, dressing flavor of sufu samples during post-fermentation was not a major factor in shaping microbial communities.

Novel Gut Microbiota Patterns Involved in the Attenuation o Dextran Sodium Sulfate-Induced Mouse Colitis Mediated by Glycerol Monolaurate via Inducing Anti-inflammatory Responses

Summary: Inflammatory bowel disease (IBD) is a chronic and relapsing inflammatory gastrointestinal condition that cannot be completely cured due to its complex pathogenesis. Glycerol monolaurate (GML) has been found to have antimicrobial, anti-inflammatory, and immunoregulatory properties. Research on mice showed that GML pretreatment was more effective in reducing weight loss, disease activity index, colonic inflammation, and proinflammatory responses compared to cotreatment. Furthermore, the increase in beneficial gut bacteria and fecal short-chain fatty acids after GML pretreatment contributed to faster and significant remission of colitis.

Differential modulations of lauric acid and its glycerides on high fat diet-induced metabolic disorders and gut microbiota dysbiosis

Summary: This study evaluated the regulation of lauric acid (LA), glycerol monolaurate (GML), and lauric triglyceride (GTL) on metabolic health and gut microbiota in high fat diet-fed mice. The results showed that LA and its glycerides improved hyperlipidemia, while GML had a more pronounced effect on glucose metabolism and inflammation. Additionally, LA and its glycerides modulated gut microbiota with increased Bifidobacterium and decreased Desulfovibrio, which were closely related to metabolic improvements.

This study found that different doses of GMD had inconsistent effects on lipid metabolism by differentially altering the gut microbiota composition. However, all doses of GMD showed excellent effects on increasing insulin sensitivity and improving inflammation.
