Extracellular vesicles from rat-bone-marrow mesenchymal stromal/stem cells improve tendon repair in rat Achilles tendon injury model in dose-dependent manner: A pilot study

Mesenchymal stromal/stem cells (MSCs) are increasingly employed for tissue regeneration, largely mediated through paracrine actions. Currently, extracellular vesicles (EVs) released by MSCs are major mediators of these paracrine effects. We evaluated whether rat-bonemarrow-MSC-derived EVs (rBMSCs-EVs) can ameliorate tendon injury in an in vivo rat model. Pro-collagen1A2 and MMP14 protein are expressed in rBMSC-EVs, and are important factors for extracellular-matrix tendon-remodeling. In addition, we found pro-collagen1A2 in rBMSC-EV surface-membranes by dot blot. In vitro on cells isolated from Achilles tendons, utilized as rBMSC-EVs recipient cells, EVs at both low and high doses induce migration of tenocytes; at higher concentration, they induce proliferation and increase expression of Collagen type I in tenocytes. Pretreatment with trypsin abrogate the effect of EVs on cell proliferation and migration, and the expression of collagen I. When either low-or high-dose rBMSCs-EVs were injected into a rat-Achilles tendon injury-model (immediately after damage), at 30 days, rBMSC-EVs were found to have accelerated the remodeling stage of tendon repair in a dose-dependent manner. At histology and histomorphology evaluation, high doses of rBMSCs-EVs produced better restoration of tendon architecture, with optimal tendon-fiber alignment and lower vascularity. Higher EV-concentrations demonstrated greater expression of collagen type I and lower expression of collagen type III. BMSC-EVs hold promise as a novel cell-free modality for the management of tendon injuries.

Return to sport after arthroscopic surgery for femoroacetabular impingement

Summary: This study analyzed the rate of return to sport, related influencing factors, and sport activity level in patients with femoroacetabular impingement (FAI) using the Hip Outcome Score - Sport-Specific Subscale (HOS-SSS). The results showed that 88.75% of patients returned to sports within a mean of 37.4 months, and post-operative sport activity level was positively associated with lighter weight, younger age, and higher preoperative scores.

Arthroscopic versus mini-open rotator cuff repair: A meta-analysis

Local Tendon Transfers for Chronic Ruptures of the Achilles Tendon: A Systematic Review

Tendon appearance at imaging may be altered, but it may not indicate pathology

Summary: In both tendon repair and tendinopathy, symptoms may not align with the appearance of the affected tendon on imaging, suggesting a dissociation between morphology and symptoms. Asymptomatic abnormal findings have limited predictive value, and interventions based on imaging are inappropriate. Patient education and clinical monitoring are recommended. Planning interventions based on sonographic abnormalities in symptomatic tendinopathy may not be warranted. Current evidence is limited and caution is needed when interpreting incidental findings in athletes.

Surface functionalization of extracellular vesicle nanoparticles with antibodies: a first study on the protein corona variable

This work explores and compares the functionalization of EVs with specific antibodies using chemisorption and physisorption methods, and reveals that the chemisorption method results in superior binding and uptake towards target cells, likely due to the stronger attachment.

Multiparametric Orthogonal Characterization of Extracellular Vesicles by Liquid Chromatography Combined with In-Line Light Scattering and Fluorescence Detection

Summary: This study demonstrates that liquid chromatography combined with multi-angle light scattering (MALS) and fluorescence detection can provide multiparametric characterization of extracellular vesicles (EVs) in one single apparatus. The method allows for analysis of EV concentration, average diameter, protein amount to particle number ratio, presence of surface markers and lipids, EV shape, and sample purity. It is fully automatable and can be applied to both crude and purified samples with limited sample handling and data analysis time.
