Effect of Novel Pyrrolo[3,4-d]pyridazinone Derivatives on Lipopolysaccharide-Induced Neuroinflammation

Neuroinflammation is considered to be one of the potential causes for the development of neurodegenerative diseases, including Alzheimer's disease. In this study, we evaluated the effect of four newly synthesized pyrrolo[3,4-d]pyridazinone derivatives on the neuron-like PC12 cells under simulated inflammation conditions by preincubation with lipopolysaccharide (LPS). Our novel derivatives are selective cyclooxygenase-2 (COX-2) inhibitors and have similar effects to nonsteroidal anti-inflammatory drugs (NSAIDs). We assessed viability (LDH assay), metabolic activity (MTT assay), DNA damage (number of double-strand breaks measured by fast halo assay), and the neuronal features of cells (average neurite length and neurite outgrowth measured spectrofluorimetrically). DCF-DA and Griess assays were also performed, which allowed determining the impact of the tested compounds on the level of oxygen free radicals and nitrites. LPS administration significantly negatively affected the results in all tests performed, and treatment with the tested derivatives in most cases significantly reduced this negative impact. Multiple-criteria decision analysis indicated that overall, the best results were observed for compounds 2a and 2b at a concentration of 10 mu M. The new derivatives showed intense activity against free oxygen radicals and nitrites. Reduced reactive oxygen species level also correlated with a decrease in the number of DNA damage. The compounds improved neuronal features, such as neurite length and outgrowth, and they also increased cell viability and mitochondrial activity. Our results suggest that derivatives 2a and 2b may also act additionally on mechanisms other than 3a and 3b.

Summary: This study aimed to determine the effect of Cornelian cherry extract on the markers important in the progress of inflammation and atherosclerotic lesion development. It found that the extract at a dose of 50 mg/kg bw resulted in a significant reduction in inflammation markers and serum levels related to atherosclerosis. The results suggest the potential usefulness of Cornelian cherry extract in preventing or treating atherosclerosis-related cardiovascular diseases.

Summary: Schizophrenia is a severe mental disorder caused by gene-environment interactions. Sex hormones, particularly testosterone, may play a role in the development and clinical features of male schizophrenic patients. Altered testosterone levels have been observed in men with schizophrenia, but the relationship between testosterone and clinical symptoms is complex and contradictory. Factors such as the phase of the disease, specific symptoms, and antipsychotic treatment may influence testosterone levels. Various factors, including illness, stress, medication, obesity, substance abuse, and liver damage, can contribute to disruptions in testosterone levels.

Cornus mas L. Extract Targets the Specific Molecules of the Th17/Treg Developmental Pathway in TNBS-Induced Experimental Colitis in Rats

Summary: Research suggests that cornelian cherry extract, rich in iridoids and polyphenols, has potential anti-inflammatory properties and can target specific molecules involved in the development of Th17/Treg cells. In an experimental colitis model, a dose of 100 mg/kg of cornelian cherry extract was effective in reducing inflammation and modulating immune factors related to Th17/Treg axis regulation. When administered with sulfasalazine, cornelian cherry extract showed even greater efficacy in reversing the pathological changes induced by colitis.

Does Inflammation Play a Major Role in the Pathogenesis of Alzheimer's Disease?

Summary: Alzheimer's disease is a neurodegenerative disease that leads to dementia, with no effective medicine currently available. The disease is characterized by the accumulation of proteins and inflammation in the brain, resulting in neuronal death. Neuroinflammation, often ignored in AD research, has gained attention recently, although the impact of comorbidities or gender differences remains unclear. This publication critically analyzes the role of inflammation in the progression of AD based on in vitro studies and other researchers' findings.

Evaluation of Recovery Methods for Fragaria vesca L. Oil: Characteristics, Stability and Bioactive Potential

Summary: Wild strawberry seed oil (WSO) recovered by cold pressing and extraction with supercritical carbon dioxide (SCO2E) was characterized for its composition and quality. The cytotoxicity of WSOs was assessed using a normal human dermal fibroblast (NHDF) cell line. Results showed that SCO2E-recovered WSO had higher tocopherol and total polyphenol contents compared to cold pressed oil. Carotenoids and squalene were highest in cold pressed oil. Phytosterol content reached 5396 mg/kg in WSO collected during 30 min of SCO2E. The highest oxidative stability was also found in this oil. All WSOs were non-cytotoxic in LDH leaching and SRB assays, but cytotoxic in the MTT test at certain concentrations for SCO2E oil collected in 15 and 30 min. The composition of WSO indicates different bio-potencies and applications depending on the extraction method.

Imatinib-Functionalized Galactose Hydrogels Loaded with Nanohydroxyapatite as a Drug Delivery System for Osteosarcoma: In Vitro Studies

Summary: This study investigates the impact of structure (XRPD, FT-IR) and surface morphology (SEM-EDS) of imatinib-functionalized galactose hydrogels, loaded and unloaded with nHAp, on osteosarcoma cell viability, levels of free oxygen radicals, and nitric oxide, levels of BCL-2, p53, and caspase 3 and 9, as well as glycoprotein-P activity. The study also examines how the rough surface of crystalline hydroxyapatite-modified hydrogels affects the release of amorphous imatinib (IM). The administration of IM and hydrogel composites shows potential in reducing the development of multidrug resistance by inhibiting P-gp.

Summary: In this study, a new series of NAH derivatives were synthesized and evaluated for their biological and physicochemical properties. Oils from Transgenic Flax Lines as Potential Chemopreventive Agents in Colorectal Cancer

Summary: This study evaluated the potential of genetically modified flaxseed oils in preventing colorectal cancer. The results showed that flaxseed oil B exhibited the strongest anti-proliferative properties, and M and B oils enhanced accumulation of Rhodamine 123 and increased colorectal cancer cell apoptosis. M oil had the highest levels of p53 protein. These transgenic oils showed utility as adjuvants in colorectal cancer treatment, specifically targeting the colon.
