Vitamin D and Sleep Regulation: Is there a Role for Vitamin D?

Background: Vitamin D exerts multiple pleiotropic effects beyond its role in calcium-phosphate metabolism. Growing evidence suggests an association between hypovitaminosis D and sleep disorders, thus increasing the interest in the role of this vitamin in the regulatory mechanisms of the sleep-wake cycle. Objective: The study aimed to explore and summarize the current knowledge about the role of vitamin D in sleep regulation and the impact of vitamin D deficiency on sleep disorders. Methods: The main regulatory mechanisms of vitamin D on sleep are explained in this study. The literature was scanned to identify clinical trials and correlation studies showing an association between vitamin D deficiency and sleep disorders. Results: Vitamin D receptors and the enzymes that control their activation and degradation are expressed in several areas of the brain involved in sleep regulation. Vitamin D is also involved in the pathways of production of Melatonin, the hormone involved in the regulation of human circadian rhythms and sleep. Furthermore, vitamin D can affect sleep indirectly through non-specific pain disorders, correlated with alterations in sleep quality. such as restless legs syndrome and obstructive sleep apnea syndrome. Conclusion: Vitamin D has both a direct and an indirect role in the regulation of sleep. Although vitamin D deficiency has been associated to sleep disorders, there is still scant evidence to concretely support the role of vitamin D supplementation in the prevention or treatment of sleep disturbances; indeed, more intervention studies are needed to better clarify these aspects.

Summary: This study aimed to evaluate whether supplementation with medium-chain fatty acids (MCTs) during a very low-calorie ketogenic diet (VLCKD) can enhance weight reduction. The study found that MCTs supplementation significantly decreased body weight, fat mass, and waist circumference, and reduced inflammation. Starting MCTs supplementation a few days before VLCKD showed the best results.

Summary: The aim of this review is to provide an overview of the menopause-related changes in microbiota and their role in the pathogenesis of menopause-related diseases. In addition, evidence on probiotic supplementation as a therapeutic strategy is discussed. Menopause is associated with dysbiosis, and these changes in the composition of microbiota in different sites (gut, vaginal, and oral microbiota) might play a role in the pathogenesis of menopause-related diseases such as osteoporosis, breast cancer, endometrial hyperplasia, periodontitis, and cardiometabolic diseases.

Liraglutide and polycystic ovary syndrome: is it only a matter of body weight?

Summary: Despite the widespread agreement on the lack of pharmacological options for the treatment of Polycystic Ovary Syndrome (PCOS), studies have shown that GLP-1 receptor agonists (GLP-1RA) could be effective in managing the clinical manifestations of PCOS, including weight loss and insulin resistance. However, current studies primarily focus on PCOS patients with obesity, and more research is needed on the lean phenotype and higher doses of GLP-1RA. Furthermore, the potential effects of GLP-1 beyond weight loss still require further investigation, especially in PCOS patients without obesity.

The quality of fats is important, and supplementation with antioxidants or micronutrients is not recommended.

Summary: Visceral obesity is associated with the progression of NASH. CK18 epitopes M30 and M65 are accurate markers for detecting NASH. This study found that CK18M30 and CK18M65 levels were significantly correlated with anthropometric and metabolic characteristics, liver stiffness, and indices of steatosis and fibrosis in subjects with visceral obesity. These levels could serve as good biomarkers for early identification of NASH.
