Noncalcemic Vitamin D Hydroxyderivatives Inhibit Human Oral Squamous Cell Carcinoma and Down-regulate Hedgehog and WNT/β-Catenin Pathways

Background/Aim: The hormonally-active form of vitamin D, 1,25(OH)(2)D-3, demonstrated activity against oral squamous cell carcinoma (OSCC). Cytochrome P450scc (CYP11A1)-derived vitamin D hydroxyderivatives, such as 20(OH)D-3 and 1,20(OH)(2)D-3, have overlapping beneficial effects with 1,25(OH)(2)D-3 without causing hypercalcemia. This study sought to determine (i) whether 20(OH)D-3 and 1,20(OH)(2)D-3 exhibit antitumor effects against OSCC comparable to those of 1,25(OH)(2)D-3 and (ii) whether these effects may stem from down-regulation of sonic hedgehog (SHH) or WNT/beta-catenin signaling pathways. Materials and Methods: Effects on CAL-27 cells were assessed by 3-(4,5dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4sulfophenyl)-2H-tetrazolium inner salt and spheroid assays. Signaling pathways were assessed by immunofluorescence and western blotting. Results: 20(OH)D-3 and 1,20(OH)(2)D-3 inhibited the growth of CAL-27 and demonstrated inhibition of WNT/beta-catenin and the SHH signaling as evidenced by down-regulation of nuclear translocation of glioma-associated oncogene 1(GLI1) and beta-catenin. Conclusion: Noncalcemic vitamin D hydroxyderivatives demonstrated antitumor activities against OSCC comparable to those of 1,25(OH)(2)D-3. Their activities against SHH and the WNT/beta-catenin pathways provide insight for a possible target for OSCC treatment.

Summary: This study established new routes for the chemical synthesis of vitamin D-3 and its hydroxy metabolites. The compounds showed similar properties in inhibiting cell proliferation and inducing cell differentiation, as well as effects on T-cell immune activity. They also interacted with nuclear receptors in a similar manner. The findings have important implications for studying the role of these compounds in the immune system and inflammatory diseases.

Molecular and structural basis of interactions of vitamin D3 hydroxyderivatives with aryl hydrocarbon receptor (AhR): An integrated experimental and computational study

Summary: This study used molecular simulation to investigate the interaction between vitamin D3 hydroxyderivatives and AhR. It was found that these derivatives can stimulate downstream target genes of AhR. Molecular dynamics simulations showed favorable binding of the derivatives to AhR and revealed potential mechanisms underlying their binding affinities.

Summary: CYP11A1 and CYP27A1 hydroxylate tachysterol(3), a photoproduct of previtamin D-3, producing 20S-hydroxytachysterol(3) and 25(OH)T-3, which are biologically active and interact with VDR, AhR, LXRs, and PPAR gamma. These metabolites inhibit cell proliferation and stimulate differentiation and anti-oxidative gene expression in keratinocytes and fibroblasts.

Vemurafenib and Dabrafenib Downregulates RIPK4 Level

Summary: RIPK4 kinase has oncogenic functions in melanoma and shows high similarity to BRAF protein, making this kinase sensitive to the BRAF inhibitors vemurafenib and dabrafenib. BRAFi downregulates RIPK4 in both BRAF-mutated and wild-type cells. Although downregulation and upregulation of RIPK4 does not affect signal transduction through the BRAF/MEK/ERK pathway in melanoma, it can inhibit cell proliferation and the FAK/AKT pathway.

Analysis of the ability of vitamin D3-metabolizing cytochromes P450 to act on vitamin D3 sulfate and 25-hydroxyvitamin D3 3-sulfate

Summary: Our study found that 25-hydroxyvitamin D3 (25(OH)D3) can exist in the human circulation in the sulfated form, and can be metabolized by vitamin D-metabolizing cytochromes P450 (CYPs). Specifically, CYP27A1 can convert vitamin D3 sulfate to 25(OH)D3 3-sulfate, while CYP11A1 can convert vitamin D3 sulfate to 20(OH)D3 3-sulfate, but at a lower rate.

Attenuation of Hypothyroidism-Induced Cognitive Impairment by Modulating Serotonin Mediation

Summary: This study aimed to examine the changes in behavior, cognition, and memory in rats with PTU-induced overt hypothyroidism and the effects of tryptophan treatment on this state. The results showed significant differences in the behavioral patterns of the hypothyroid rats, including reduced locomotor activity, rearing, and memory function, which were alleviated by treatment with 5-OH-TRP. These findings have important implications for further investigation of functional and structural changes in the hippocampus during a hypothyroid state and the effects of serotonin mediation on hypothyroid-associated depressive behavior.

Inactivation of vitamin D2 metabolites by human CYP24A1

Summary: Vitamin D exists in two forms, D3 produced in the skin and D2 obtained from the diet. The enzyme CYP24A1 plays a crucial role in oxidizing and inactivating both forms of vitamin D, regulating their serum levels. This study aimed to clarify the pathways of oxidation for 25(OH)D2 and 1,25(OH)(2)D2 and compare their metabolism with their vitamin D3 counterparts.

Metabolism of Lumisterol2 by CYP27A1

Summary: Lumisterol2 (L2) is a photoproduct of UVB action on ergosterol, which is present in edible mushrooms and human skin. CYP27A1 efficiently metabolizes L2 to produce three major hydroxy-L2 products, while CYP11A1 has minimal effect on L2 metabolism. The major CYP27A1-derived hydroxy-L2 products have anti-proliferative activity on skin-cancer cell lines.
